Membrane Potential

Membrane potential is the voltage difference across a cell membrane, created by unequal ion distributions, and it enables neurons to receive, process, and transmit information. In neurons, the sodium-potassium pump establishes concentration gradients, while selective ion channels regulate permeability and determine the membrane’s voltage; changes in these channels can produce depolarization, repolarization, and action potentials. Studying membrane potential helps explain neuronal signaling, synaptic communication, sensory processing, and muscle activation. Experimental measurements of these voltage changes also support research into ion channel function, neurological disorders, and the effects of drugs that alter electrical excitability.

The Resting Membrane Potential

2019

Overview The relative difference in electrical charge, or voltage, between the inside and the outside of a cell membrane, is called the membrane potential. It is generated by differences in permeability of the membrane to various ions and the concentrations of these ions across the membrane. The Inside of a Neuron Is More Negative The membrane potential of a cell can be measured by inserting a microelectrode into a cell and comparing the charge to a reference electrode in the extracellular...

Source: Hakim, M. A. et al., Isolation and Functional Analysis of Arteriolar Endothelium of Mouse Brain Parenchyma. J. Vis. Exp. (2022).This video demonstrates the functional analysis of an arteriolar endothelial tube isolated from a mouse brain. It outlines the steps for measuring intracellular calcium levels and membrane potential, both at baseline and in response to pharmacological stimulation.

Simultaneous Measurement of Intracellular Calcium and Membrane Potential in a Mouse Cerebral Endothelium

2025

The video demonstrates the simultaneous measurement of intracellular calcium levels and changes in membrane potential in endothelial cells of mouse cerebral artery segments. The isolated endothelial tube is loaded with a fluorescent calcium indicator and treated with a drug that induces intracellular calcium release, resulting in a change in membrane potential. The indicator's fluorescence enables the measurement of calcium levels, and a recording electrode is inserted into a cell to measure...
