Spike Response Recording

Spike response recording is a neuroscience technique that measures the electrical action potentials, or spikes, produced by neurons in response to stimuli or experimental conditions. Recording electrodes detect rapid voltage changes associated with membrane depolarization, while amplification and signal processing distinguish individual spikes and quantify their timing, frequency, and patterns. These measurements reveal how neurons encode sensory information, communicate within circuits, and change their excitability during development, disease, or drug exposure. Spike response recordings therefore support studies of neural coding, synaptic function, and circuit activity, linking cellular electrical behavior to broader nervous system functions.

Optical Recording of Suprathreshold Neural Activity with Single-cell and Single-spike Resolution

2012

Understanding the function of the vertebrate central nervous system requires recordings from many neurons because cortical function arises on the level of populations of neurons. Here we describe an optical method to record suprathreshold neural activity with single-cell and single-spike resolution, dithered random-access scanning. This method records somatic fluorescence calcium signals from up to 100 neurons with high temporal resolution. A maximum-likelihood algorithm deconvolves the...

Electrophysiological Recordings from Drosophila Sensilla in Response to Volatile Odorants

2025

In this video, a live Drosophila fly is mounted in a tube for electrophysiological studies. Electrodes are positioned at the base of the sensillum to record specific neuronal responses to pheromones with the fly's olfactory receptor neurons.

Recording CA3-CA1 Synaptic Responses in a Rat Hippocampal Slice

2025

This video demonstrates a method to record CA3-CA1 synaptic responses in a rat hippocampal slice. It outlines the steps involved in placing the stimulating and recording electrodes on the hippocampal slice and measuring the field excitatory postsynaptic potential (fEPSP) to assess synaptic response strength.

The overall goal of this protocol is to demonstrate how to present odorants of low volatility for single-sensillum recording from Drosophila olfactory receptor neurons that respond to long-chain cuticular pheromones.

Triggering a Closed-Loop Stimulation by Neuron Spiking Activity

2025

This video demonstrates the operation of closed-loop stimulation in a rat with a surgically implanted device to record neural activity. Neuronal signals are first recorded and analyzed. Template waveforms are then uploaded, and parameters are defined to trigger closed-loop stimulation.
