Spine Motility Analysis

Spine motility analysis is a neuroscience method for measuring the dynamic changes in shape, size, and position of dendritic spines, the small neuronal protrusions that often receive excitatory synaptic input. Using time-lapse fluorescence or live-cell microscopy, researchers track spine formation, elongation, retraction, and stabilization as actin cytoskeleton remodeling alters spine structure over time. These measurements reveal how synaptic connections develop and respond to neuronal activity, experience, or molecular signals. Spine motility analysis is therefore valuable for studying synaptic plasticity, learning and memory, and structural changes associated with neurological and psychiatric disorders.

Numerous recent studies have identified mutations in synaptic proteins associated with brain pathologies. Primary cultured cortical neurons offer great flexibility in examining the effects of these disease-associated proteins on dendritic spine morphology and motility.
