Transgenic Mice

Transgenic mice are laboratory animals whose genomes have been deliberately modified to carry an introduced gene or an engineered genetic sequence, making them valuable models for studying gene function and disease. The inserted DNA can be expressed in selected cells or controlled by specific promoters, while methods such as embryo microinjection, embryonic stem-cell targeting, or CRISPR-based editing establish heritable changes. In neuroscience, these models help link genes to neuronal development, synaptic signaling, behavior, and disorders such as Alzheimer’s disease or epilepsy. Reporter and conditional lines also allow researchers to visualize cell populations or activate or disable genes at defined times, supporting causal studies and therapeutic evaluation.

The microinjection of mouse oocytes is commonly used for both classic transgenesis (i.e., the random integration of transgenes) and CRISPR-mediated gene targeting. This protocol reviews the latest developments in microinjection, with a particular emphasis on quality control and genotyping strategies.

This protocol describes the dynamics of viral infections using luciferase- and fluorescence-expressing recombinant (r)SARS-CoV-2 and an in vivo imaging systems (IVIS) in K18 hACE2 transgenic mice to overcome the need of secondary approaches required to study SARS-CoV-2 infections in vivo.

Installation of Optical Windows in the Brain of Transgenic Mice to Image Neural Activities

2025

This video demonstrates the brain's neural activity recording through an optical window in transgenic mice. The mice express a fluorescent protein that enables the detection of neural activities. A small portion of the skull is carefully removed to install an optical window, and the skull is then covered with black cement to block light before imaging.
