In Vitro Methods

In vitro methods are laboratory approaches that examine biological or pharmacological processes outside a whole organism, using isolated cells, tissues, enzymes, or receptor systems under controlled conditions. By exposing these preparations to defined concentrations of a compound and measuring responses such as receptor binding, enzyme activity, cell viability, or signal transduction, researchers can characterize drug action and concentration-dependent effects. In pharmacology, these methods support early screening, mechanism-of-action studies, toxicity assessment, and comparisons of candidate therapies before animal or clinical testing. They reduce experimental complexity, improve control over variables, and provide quantitative evidence for selecting compounds and designing subsequent studies.

An In Vitro Method for the Differentiation of Megakaryocytes and Platelet Formation

2025

This video demonstrates a technique for the differentiation of megakaryocytes and the formation of platelets. Hematopoietic stem progenitor cells — isolated from human umbilical cord blood — differentiate into mature megakaryocytes in the presence of thrombopoietin. The mature megakaryocytes produce cytoplasmic extensions termed proplatelets, which give rise to individual platelets.

Methods for Studying Drug Absorption: In vitro

2024

In vitro experiments are crucial for understanding the transport and absorption of drugs through biological materials. These studies employ varied methods such as the diffusion cell method, the everted sac technique, and the everted ring technique. The diffusion cell method uses a two-compartment cell, including a donor compartment with the drug solution, which simulates the environment where the drug is applied, and a receptor compartment with a buffer solution, which simulates the environment...

In Vitro Drug Dissolution: Alternative Methods

Alternative drug dissolution methods include the rotating bottle, intrinsic dissolution test, peristalsis, and the Franz diffusion cell method. The rotating bottle method involves meticulously rotating tightly capped controlled-release beads in a temperature-controlled bath. Periodic decanting of samples allows for residue assay, followed by refilling with fresh medium and testing at various pH levels to emulate the gastrointestinal tract conditions. In contrast, the intrinsic dissolution test...
