Cytokine Array Analysis

Cytokine array analysis is a multiplex immunoassay that measures multiple cytokines, small signaling proteins, in a biological sample at the same time, making it useful for characterizing complex cellular communication. In a typical array, capture antibodies immobilized on a membrane or solid surface bind target cytokines; detection antibodies then produce a measurable signal, such as fluorescence or chemiluminescence, whose intensity is compared with reference standards. Quantitative analysis applies signal detection, calibration, and spatial pattern recognition, linking the technique to measurement principles in physics. It supports studies of inflammation, immune responses, disease mechanisms, and treatment effects.

Described here is a method for producing exogenous cytokine in patient-derived xenograft (PDX) mice via weekly intraperitoneal injection of a cytokine-transduced stromal cell line. This method broadens the utility of PDX and provides the option for transient or sustained exogenous cytokine delivery in a multitude of PDX models.
