Question 1

What is differential scanning calorimetry and how does it measure thermal properties?

Accepted Answer

Differential scanning calorimetry (DSC) is a measurement technique that characterizes thermal properties of materials by comparing heat flow between a sample pan and an empty reference pan. Both pans are heated identically, and the difference in energy required to maintain equal temperatures is recorded versus temperature. This heat flow data reveals phase transitions, melting events, and other thermodynamic changes in the material.

Question 2

How do crystalline, amorphous, and semi-crystalline polymers differ in their thermal behavior?

Accepted Answer

Crystalline polymers have long-range ordered chain structures and undergo melting during heating. Amorphous polymers have randomly organized chains and transition from rigid glass to rubbery states through glass transition. Semi-crystalline polymers contain both ordered and disordered regions, so they exhibit both glass transition in amorphous parts and melting in crystalline parts during DSC heating.

Question 3

What do endothermic and exothermic peaks represent in a DSC curve?

Accepted Answer

In DSC plots, endothermic events—those absorbing heat—appear as valleys or downward peaks, while exothermic events—those releasing heat—appear as upward peaks. The heat of melting (ΔHm) is calculated from the area under endothermic peaks during heating, and the heat of cold crystallization (ΔHc) is calculated from exothermic peaks during cooling, allowing percent crystallinity determination.

Question 4

How is polymer percent crystallinity calculated using DSC data?

Accepted Answer

Percent crystallinity is determined by measuring both the heat of melting (ΔHm) during heating and the heat of cold crystallization (ΔHc) during cooling phases. These values are obtained from the areas under their respective DSC peaks. Using the known heat of melting for a 100% crystalline reference sample, the percent crystallinity is calculated as a ratio of the measured values to the reference value.

Question 5

What preparation steps are required before running a DSC measurement?

Accepted Answer

Before measurement, allow the DSC instrument to warm up for about an hour and verify that compressed nitrogen and liquid nitrogen tanks are full. Prepare two chemically inert pans by poking small holes in their lids and sealing them with a crimping press. Place empty pans on furnace sensors, define measurement parameters in software including temperature program and calibration files, and verify the furnace is within five degrees of the initial temperature before starting.

Question 6

How should polymer samples be prepared and loaded into DSC pans?

Accepted Answer

Cut polymer samples into small pieces and weigh the empty pan with lid first. Arrange sample pieces in a thin, even layer covering the entire pan bottom to ensure uniform heat flow. Seal the pan with a crimped lid, then weigh the full pan and subtract the empty pan weight to determine sample mass. Input the sample name and mass into the DSC software before beginning the scan.

Question 7

What applications extend DSC beyond polymer crystallinity analysis?

Accepted Answer

DSC measures enthalpy changes during chemical reactions and phase transitions in diverse materials. For biological samples, DSC analyzes freeze-drying properties by measuring glass transitions in frozen cell suspensions, helping optimize storage conditions. DSC also quantifies decomposition reactions, such as calculating the enthalpy of calcium carbonate decomposition, making it valuable for studying thermodynamic events across materials science and biological research.
