Os exploradores europeus tinham conhecimentos, habilidades e equipamentos adequados, mas por que não chegaram ao Novo Mundo até o final do século 15? Você ficará surpreso ao saber que a resposta a essa pergunta está relacionada às pequenas moléculas biológicas chamadas enzimas, que atuam como catalisadores. 

O que é um catalisador? É uma substância que permite que uma reação ocorra mais rapidamente, usando menos energia. Se você traçasse um gráfico da energia necessária para uma reação biológica não catalisada, seria mais ou menos assim. Essa altura representa a energia de ativação, que é a quantidade mínima de energia necessária para que a reação prossiga. As enzimas, funcionando como catalisadores, fornecem uma via de reação alternativa, com menor energia de ativação. 

Então, como as enzimas realmente funcionam? A maioria das enzimas são proteínas. A parte de sua estrutura que interage com as moléculas que catalisam é chamada de sítio ativo, que tem afinidade apenas por certos substratos e não se liga a outros. Quando os substratos corretos entram no sítio ativo, a enzima muda sua forma para atingir uma configuração de maior afinidade. Esse estado de transição facilita a reação e converte os reagentes em produtos. A enzima, no entanto, tem uma afinidade menor com os produtos, por isso os libera e retorna à sua forma original, pronta para repetir o mesmo processo. Este é um exemplo de enzima catabólica porque pegou um único substrato e o quebrou para formar vários produtos. O oposto disso é a enzima anabólica, que pega dois substratos e os une para formar um produto maior. 

Existem inúmeras outras maneiras pelas quais as enzimas funcionam. Por exemplo, essa enzima transfigura um único substrato em uma nova forma. Então, você pode estar se perguntando, como tudo isso se relaciona com as viagens do século 15? Naquela época, os marinheiros não tinham acesso a produtos frescos contendo vitamina C, uma coenzima essencial para a síntese de colágeno. Por causa disso, os marinheiros desenvolveriam escorbuto, uma doença por deficiência de vitamina C, que muitas vezes era fatal. Eles resolveram esse problema conservando frutas e vegetais ricos em vitamina C. A decapagem reduziu o pH do alimento, o que impediu a atividade das enzimas microbianas, responsáveis pelo apodrecimento. Portanto, a destruição de enzimas microbianas e a economia de uma coenzima essencial desempenharam um papel importante no sucesso de longas expedições. 

Neste laboratório, você pode quantificar como diferentes condições afetam a função enzimática, avaliando a taxa de reação, a quantidade de produto produzido, por unidade de tempo. Você primeiro medirá a taxa de reação da linha de base em uma temperatura ambiente padrão e pH neutro. A taxa aumentará no início até que todas as moléculas enzimáticas estejam completamente saturadas com o substrato. E então, a catálise atingirá um nível constante, pois as enzimas estão ocupadas continuando o ciclo de substrato de ligação, catálise e liberação. Uma vez estabelecida a taxa de reação basal, ela pode ser usada como ponto de referência para outras condições. A enzima com a qual você trabalhará neste laboratório é a peroxidase de nabo. <h4>Catalisadores biológicos</h4>

Todos os organismos vivos realizam continuamente inúmeras reações bioquímicas para sustentar sua presença. A maioria dessas reações requer uma entrada de energia para começar, que é chamada de energia de ativação. Catalisadores são produtos químicos que diminuem a energia de ativação. Embora os catalisadores facilitem uma reação química, eles não são consumidos por ela. Isso significa que um catalisador pode facilitar uma reação química específica repetidamente em sucessão, aumentando a taxa dessa reação.

As enzimas são catalisadores biológicos que aumentam a velocidade e a eficiência das reações bioquímicas. A maioria das enzimas são proteínas, mas certas moléculas de ácido ribonucléico também têm propriedades catalíticas. Cada tipo de enzima só pode se ligar a um ou alguns substratos específicos, portanto, existe uma grande diversidade de enzimas, todas com funções distintas. A reação específica de uma enzima depende de seus sítios ativos, que têm formas que se aproximam da forma de seus substratos. Após a ligação ao substrato, a enzima muda ligeiramente de forma de uma forma que permite que a reação química aconteça mais prontamente, reduzindo assim a energia de ativação da reação. Uma vez que a reação ocorre, a enzima libera os produtos e retorna à sua forma original, o que permite repetir o processo novamente.

<h4>Enzimas catabolíticas versus anabólicas</h4>

A maioria das reações enzimáticas pode ser categorizada como catabólica e anabólica, que têm funções opostas. As enzimas que medeiam reações catabólicas decompõem substâncias maiores em vários produtos. Uma enzima catabólica bem conhecida é a lactase, que divide o dissacarídeo lactose em produtos lácteos em monossacarídeos galactose e glicose. Os seres humanos têm grandes quantidades de lactase no intestino delgado após o nascimento, no entanto, muitos indivíduos perdem mais de 90% de sua lactase naprimeira infância. Essa redução diminui a capacidade de digerir a lactose de maneira dependente da idade, o que é conhecido como intolerância à lactose2.

As enzimas anabólicas combinam várias substâncias em um único produto. Por exemplo, a enzima DNA polimerase sintetiza moléculas de DNA unindo nucleotídeos. Da mesma forma, a enzima glicogênio sintase conecta moléculas de glicose para gerar glicogênio. A insulina regula a atividade da glicogênio sintase, portanto, pacientes diabéticos com níveis baixos ou inexistentes de insulina não podem sintetizar glicogênio sem tratamento com insulina3.

<h4>Fatores ambientais</h4>

A forma e a carga do sítio ativo de uma enzima são adaptadas à reação química específica que ela catalisa, portanto, a atividade enzimática é influenciada por fatores que afetam seu sítio ativo. Portanto, as enzimas requerem condições específicas para funcionar em níveis ideais e, dependendo de sua função e localização, essas condições podem variar muito. Esses fatores incluem pH e temperatura. Por exemplo, sítios ativos de enzimas geralmente contêm aminoácidos ácidos ou básicos. Desvios do pH ideal alteram as cargas no sítio ativo, dificultando a interação dos substratos com a enzima. Essa propriedade tem sido usada há séculos para preservar alimentos por decapagem, o que reduz o pH do alimento em conserva a um nível que inibe efetivamente a atividade enzimática microbiana. Da mesma forma, cada enzima funciona em uma faixa de temperatura ideal. Se a temperatura mudar abaixo ou acima dessa faixa, ela altera a forma do sítio ativo, impedindo-o de interagir efetivamente com seus substratos. Na maioria dos casos, retornar à temperatura ideal restaura a forma e a função originais da enzima, no entanto, temperaturas muito altas podem desnaturar ou danificar irreversivelmente a estrutura da enzima. Extremófilos, ou organismos encontrados em condições extremas que são letais para a maioria das formas de vida, têm enzimas capazes de funcionar nesses extremos. Por exemplo, Thermus aquaticus é uma bactéria encontrada em fontes termais e fontes hidrotermais que atingem temperaturas nas quais a maioria das proteínas se desnaturaria. A taq polimerase é uma DNA polimerase termoestável encontrada em T. aquaticus, que permitiu o desenvolvimento da reação em cadeia da polimerase (PCR), um método laboratorial amplamente utilizado para replicação rápida do DNA. A PCR requer altas temperaturas para separar as fitas da dupla hélice do DNA a serem replicadas. A Taq polimerase permanece estável nessas altas temperaturas, enquanto a maioria das enzimas seria degradada. A PCR alterna entre altas temperaturas que quebram o DNA em fitas simples e temperaturas mais baixas que permitem que a polimerase construa um novo DNA de fita dupla.

<h4>Fatores moleculares que influenciam a atividade enzimática</h4>

Além de faixas específicas de pH e temperatura, algumas enzimas funcionam melhor na presença de outras moléculas ou íons. As coenzimas são biomoléculas que se ligam ao sítio ativo da enzima e participam da catálise sem serem consumidas pela reação. As coenzimas geralmente transportam elétrons, átomos específicos e grupos funcionais e, portanto, atuam como transportadores intermediários. As vitaminas B atuam como coenzimas para inúmeras reações bioquímicas. Por exemplo, a coenzima A (coA), que é a forma ativa do ácido pantotênico, é necessária em 4% de todas as enzimas de mamíferos4. Os cofatores são frequentemente classificados como substâncias inorgânicas, como íons, que são necessárias para a função enzimática. Por exemplo, todas as enzimas DNA polimerase requerem Mg2+ ou Mn2+ para replicação do DNA, bem como excisão de nucleotídeos incorporados incorretamente5.

Existem também outras moléculas que não são necessárias para o funcionamento de uma enzima, mas são capazes de alterar a atividade enzimática. As substâncias que aumentam a atividade enzimática são chamadas de ativadores, enquanto outras que diminuem a atividade enzimática são chamadas de inibidores. Essas moléculas se ligam a enzimas no sítio ativo ou em outros locais da enzima conhecidos como sítios alostéricos. Às vezes, essas moléculas podem se ligar ao local exato no sítio ativo como substrato e, assim, competir com o substrato pela ligação enzimática. Outros, especialmente as substâncias que se ligam a sítios alostéricos, podem alterar a função enzimática sem bloquear a ligação do substrato ao sítio ativo e, portanto, não são competitivas. Além disso, a ligação dos inibidores pode ser reversível ou irreversível. Quando a interação é reversível, a enzima pode retomar sua função assim que o inibidor se desprende. No entanto, quando a ligação é irreversível, o inibidor nunca se desprende da enzima e a enzima fica irreparavelmente danificada e precisa ser substituída. No entanto, os inibidores enzimáticos também são úteis. Por exemplo, os inibidores têm sido utilizados como antibióticos para inibir o crescimento bacteriano e como quimioterápicos para inibir a divisão descontrolada das células cancerígenas6.

<h4>Medindo a função enzimática</h4>

Um método usado pelos cientistas para estudar os efeitos de diferentes condições na função enzimática é quantificar a taxa de reação, que é a rapidez com que a enzima está catalisando a reação. Portanto, uma taxa de reação mais alta indica que a enzima está catalisando reações com mais eficiência, enquanto uma taxa mais baixa significa que ela está funcionando com menos eficiência. Para determinar a taxa de reação, a velocidade de reação é calculada medindo a quantidade de produto produzido por unidade de tempo. Isso é feito medindo a concentração do produto começando logo no início da reação, quando a concentração do produto está aumentando linearmente, até que a enzima esteja completamente saturada com o substrato e a catálise atinja um nível constante. Uma vez determinada a taxa de reação basal, ela pode ser usada como ponto de referência, ou controle, ao comparar entre uma variedade de tratamentos. Podemos avaliar as taxas de reação como medidas relativas ou absolutas. Uma taxa de reação relativa é uma medida comparativa que mostra diferenças entre duas ou mais condições, como tratamentos em um único experimento, mas não fornece um valor para o que está sendo medido. Uma taxa de reação absoluta depende de uma medida quantitativa do progresso da reação. Uma maneira fácil de entender a diferença entre as taxas de reação relativas e absolutas é considerar a medição da velocidade de um carro em uma corrida. Um método relativo seria comparado avaliando a classificação dos carros na linha de chegada, enquanto a taxa absoluta avaliaria e compararia a velocidade de carros individuais em milhas por hora.

Como as enzimas são tão amplamente utilizadas, os pesquisadores começaram a procurar maneiras de criar e modificar enzimas para atender a necessidades específicas, conhecidas como engenharia de proteínas. A tecnologia de DNA recombinante permite que as sequências de aminoácidos sejam alteradas para alterar a forma de uma enzima ou a afinidade do sítio ativo. Isso tem sido usado para criar testes de diagnóstico altamente sensíveis para várias condições, bem como para esforços de biorremediação e ferramentas de gerenciamento de resíduos7.

Enzymes are biological catalysts that help living things run biochemical reactions faster. They lower activation energy, which is the energy needed to start a reaction. Enzymes are not used up in the process, so they can keep working again and again. Most enzymes are proteins, although some ribonucleic acid molecules also have catalytic activity.

Each enzyme usually binds only one or a few specific substrates. This selectivity depends on the enzyme’s active site, which fits its substrate like a matching shape. When the substrate binds, the enzyme changes shape slightly and makes the reaction easier. After the products form, the enzyme releases them and returns to its original shape.

Enzyme reactions are often grouped as catabolic or anabolic. Catabolic enzymes break larger molecules into smaller products. Lactase is a catabolic enzyme that splits lactose in dairy products into galactose and glucose. Many people lose most of their lactase by early childhood, which can lead to lactose intolerance. Anabolic enzymes build larger molecules from smaller parts. DNA polymerase joins nucleotides to make DNA, and glycogen synthase links glucose molecules to form glycogen. Insulin helps regulate glycogen synthase, so people with little or no insulin may need treatment to make glycogen normally.

Enzyme activity depends on conditions such as pH and temperature. Many active sites contain acidic or basic amino acids, so changes in pH can alter charge and make substrate binding harder. Pickling uses this idea by lowering pH and slowing microbial enzyme activity. Temperature also matters because each enzyme has an optimal range. Low or moderate shifts may slow the enzyme, but high temperatures can denature it and cause lasting damage. Cooking meat and refrigerating food both reduce harmful microbial activity in this way. Extremophiles live in harsh environments and have enzymes that still work there. Thermus aquaticus is one example, and its Taq polymerase stays stable in the high heat used for PCR, a method that copies DNA by cycling between hot and cooler temperatures.

Some enzymes also need coenzymes or cofactors. Coenzymes are biomolecules that help the reaction and are not consumed. They often carry electrons, atoms, or functional groups, and many B vitamins serve this role. Coenzyme A, the active form of pantothenic acid, is needed by many mammalian enzymes. Cofactors are usually inorganic substances, such as ions. DNA polymerase, for example, requires Mg2+ or Mn2+ for DNA replication and for correcting some mistakes.

Other molecules can change enzyme activity without being required for the reaction. Activators increase activity, while inhibitors decrease it. These molecules may bind at the active site or at allosteric sites, which are different locations on the enzyme. Some inhibitors compete with the substrate for the active site, while others act in a non-competitive way by binding elsewhere. Inhibitor binding can also be reversible or irreversible. Reversible inhibitors can detach and let the enzyme work again, but irreversible inhibitors permanently damage the enzyme. Scientists use enzyme inhibitors as antibiotics and as chemotherapeutic agents to limit cancer cell division.

Scientists measure enzyme function by looking at reaction rate, or how fast product is made. A faster rate means the enzyme is working more efficiently. Reaction velocity is measured by tracking product concentration over time, usually from the start of the reaction until the enzyme becomes saturated with substrate and the rate levels off. Researchers use that baseline as a control when comparing different treatments. Reaction rates can be reported as relative or absolute values. Relative rates compare conditions, while absolute rates give a direct quantitative measure, much like comparing race rankings versus miles per hour.

Because enzymes are so useful, researchers also engineer them for special tasks. Protein engineering uses recombinant DNA technology to change amino acid sequences and adjust enzyme shape or active site binding. This approach has helped create sensitive diagnostic tests, as well as tools for bioremediation and waste management.
