Cell Structure

A vida, desde os maiores organismos até os mais minúsculos, é composta por células. As células são os blocos de construção da vida e fornecem os materiais necessários para a existência da vida. 
Embora as células de todas as esferas da vida contenham estruturas semelhantes, como DNA e proteína, existem dois tipos diferentes de células com base em como essas estruturas são organizadas internamente. As células procarióticas, as de bactérias e arqueias, são simples em sua organização. Por exemplo, embora seu genoma esteja densamente compactado, ele não é cercado por uma membrana. As células eucarióticas, aquelas que compõem animais, plantas e fungos, são mais complexas. Eles compartimentam suas estruturas internas em organelas, estruturas ligadas à membrana que desempenham funções específicas. Em contraste com os procariontes, o DNA eucariótico é encerrado em uma membrana nuclear. Outras organelas incluem o retículo endoplasmático, a mitocôndria e o lisossomo.

A célula vegetal também inclui um grande vacúolo para armazenar proteínas, metabólitos e água. Por fim, as células vegetais também possuem uma parede celular rígida, necessária para o crescimento e retenção de água. 
Como as células são tão pequenas, elas só podem ser vistas por microscopia. Uma das maneiras mais simples de visualizar as células é através do microscópio de campo claro. Este microscópio contém uma fonte de luz, uma lente condensadora para focalizar a luz, uma lente ocular para visualizar a amostra e uma lente objetiva para ampliação. Um microscópio não é a única coisa necessária para visualizar o invisível. As estruturas dentro das células são transparentes, então as células devem ser coradas com corantes. Por exemplo, a safranina é usada para corar os núcleos das células de vermelho.

<h4>Fundo</h4>
As células representam as unidades biológicas mais básicas de todos os organismos, sejam organismos unicelulares simples, como bactérias, ou grandes organismos multicelulares, como elefantes e sequoias gigantes. Em meados doséculo 19, a Teoria Celular foi proposta para definir uma célula, que afirma:

	<li>Todo organismo vivo é composto de uma ou mais células.</li>
	<li>As células são as unidades funcionais de todos os organismos.</li>
	<li>Todas as células surgem de células preexistentes.</li>
</ul>
Todas as células compartilham características comuns, como ter uma membrana plasmática, um citoplasma, DNA e ribossomos. Uma membrana plasmática é uma bicamada fosfolipídica que envolve a célula. Essa camada fina e fluida ao redor das células serve para isolar o conteúdo da célula de seu ambiente e regula a troca de material com seu ambiente, além de facilitar as interações com outras células. Dentro da membrana plasmática, a célula é preenchida com um fluido semelhante a um gel chamado citoplasma, que contém moléculas orgânicas, sais e outros materiais vitais para as funções da célula. Portanto, as reações bioquímicas que sustentam a vida, conhecidas como processos metabólicos, ocorrem dentro do citoplasma. Os tipos de processos metabólicos que uma célula pode executar dependem de sua informação genética. Todas as células usam o DNA como material genético, que é o modelo hereditário para construir estruturas e produtos celulares. Finalmente, todas as células usam ribossomos para sintetizar seus produtos proteicos.
Existem dois tipos de células com base na localização de seu material genético: procarióticas, que significa "antes do núcleo", e eucarióticas, que significam "núcleo verdadeiro". Portanto, embora ambos os tipos de organismos tenham DNA, procariontes como bactérias têm nucleóides ou componentes "semelhantes a núcleos" em vez de um núcleo, enquanto os eucariotos possuem núcleos verdadeiros ligados à membrana para conter seu DNA. Além disso, os procariontes são relativamente pequenos, em torno de 0,1 a 5,0 micrômetros (μm), em comparação com os eucariotos, que normalmente podem variar de 10 a 100 μm de tamanho. As bactérias engolfadas evoluíram para as primeiras organelas produtoras de energia, as mitocôndrias, que são as organelas do metabolismo aeróbico da célula. As mitocôndrias têm seu próprio genoma separado e são semelhantes em tamanho aos procariontes. Eles contêm duas camadas de membranas que envolvem dois compartimentos distintos. Algumas das reações que quebram as biomoléculas de alta energia ocorrem no compartimento interno, enquanto o compartimento externo abriga as reações que capturam a energia liberada desses compostos em moléculas de trifosfato de adenosina (ATP) para serem usadas como moeda de energia da célula.
Núcleos e mitocôndrias não são as únicas estruturas compartilhadas de células eucarióticas. Outras organelas eucarióticas onipresentes são o retículo endoplasmático liso e rugoso (RE), aparelho de Golgi, lisossomos e vacúolos. Retículo endoplasmático significa simplesmente "rede dentro do plasma" e, como o próprio nome indica, é uma grande rede de membranas dentro da célula, especialmente ao redor do núcleo. Partes do RE rugoso se estendem da membrana nuclear e se distinguem do RE liso por sua aparência áspera devido a numerosos ribossomos em sua superfície. O ER rugoso é o local para a síntese de proteínas, como as proteínas que estão embutidas na membrana plasmática ou as proteínas que são secretadas pela célula. Em contraste, o ER liso produz produtos à base de lipídios, mas também contém enzimas para desintoxicar produtos químicos nocivos. Portanto, as células do fígado contêm ER liso abundante. Além disso, as células musculares contêm quantidades significativas de ER liso devido à função de armazenamento de cálcio dessa organela, que é essencial para a contração muscular. O aparelho de Golgi classifica, modifica e empacota produtos celulares dentro de vesículas, que se fundem com a membrana plasmática para liberar os produtos. Algumas das proteínas produzidas no RE rugoso são enzimas digestivas intracelulares. Essas enzimas são empacotadas no aparelho de Golgi em vesículas especiais chamadas lisossomos. A principal função dos lisossomos é digerir as partículas de alimentos engolidas pela célula, bem como as partes das células velhas. Os vacúolos são sacos de membrana celular que servem como unidades de armazenamento dentro das células. Eles podem servir para armazenar água para regular o conteúdo de água da célula, bem como armazenar produtos metabólicos ou mesmo moléculas venenosas, dependendo do tipo de célula e do organismo.
<h4>Organelas específicas do reino</h4>
As células eucarióticas também desenvolveram organelas distintas, específicas para cada reino. Por exemplo, o reino Plantae e Animalia são ambos eucarióticos, no entanto, as organelas das células vegetais e animais diferem de maneiras importantes que lhes permitem levar suas vidas como produtores e consumidores, respectivamente. As plantas terrestres precisam crescer altas e ter caules rígidos para segurar as folhas, que usam para a fotossíntese. Eles também precisam ser capazes de reter a água absorvida pelas raízes. Suas células refletem essas necessidades específicas. Ao contrário das células animais, as células vegetais têm cloroplastos, que são usados para a fotossíntese e geralmente contêm o pigmento verde clorofila. Além disso, eles são cercados por paredes celulares, que são camadas externas rígidas feitas de celulose para apoiar o crescimento e a retenção de água. Como eles precisam armazenar grandes quantidades de água para manter a pressão da água na célula, eles têm vacúolos maiores do que as células animais. Além disso, as células vegetais também possuem outro tipo de organelas de armazenamento especializadas chamadas plastídios, que contêm pigmentos e produtos fotossintéticos, como o amido. Essas diferenças são perceptíveis e distinguem as células vegetais das células animais: as células vegetais geralmente têm uma forma regular e retangular devido às suas paredes celulares rígidas, enquanto as células animais são arredondadas e mais irregulares.
<h4>Microscopia</h4>
Algumas células, como oócitos de rã, são grandes o suficiente para serem vistas a olho nu, mas a maioria das células não pode ser vista sem qualquer auxílio visual. Portanto, os cientistas usam técnicas de microscopia para estudar as estruturas celulares e distinguir os tipos de células uns dos outros. Embora os microscópios sejam capazes de ampliar objetos difíceis ou impossíveis de serem vistos com o olho humano, a maioria dos tecidos naturalmente carece de pigmentação. Portanto, foram criadas soluções que podem corar seletivamente as células com base em sua composição molecular. Isso permite que os pesquisadores distingam entre organelas em uma célula, tipos de tecido em um caule vegetal e camadas de gordura em animais, apenas para citar alguns exemplos. O corante azul de metileno cora os ácidos nucléicos das células mortas, ligando-se ao DNA carregado negativamente. A solução de safranina é outro corante biológico que mancha os núcleos das células de vermelho. As células só precisam estar nas soluções de coloração por um curto período de tempo e podem ser montadas imediatamente após a etapa de coloração. As técnicas de montagem comumente usadas são montagem úmida e imersão em óleo. Uma montagem úmida é criada coletando uma amostra e colocando-a em uma lâmina de vidro com líquido entre a lâmina e a lamínula. As amostras de células são suspensas em líquidos como água ou glicerol. O glicerol é melhor para usar com culturas vivas, pois evita a proliferação de bactérias3. O óleo de imersão pode ser adicionado em cima da lamínula para melhorar a visualização da amostra em alta ampliação. Isso é feito porque o óleo tem o mesmo índice de refração que o vidro, o que significa que permite que a luz passe por ele tão bem quanto o vidro. A interface vidro-ar espalha a luz mais do que o óleo ou o vidro, de modo que a clareza da imagem é afetada quando as amostras são montadas "secas" ou sem óleo. Uma vez que as células são coradas e montadas, elas estão prontas para serem estudadas ao microscópio.
Existem várias técnicas de microscopia, desde a tecnologia de varredura eletrônica que permitiu aos pesquisadores visualizar objetos em nível atômico até imagens fluorescentes de células vivas que permitem o monitoramento em tempo real do movimento de moléculas dentro de células individuais4. A microscopia de campo claro é a técnica de microscopia mais simples, exigindo apenas uma fonte de luz halógena, uma lente condensadora para focar a luz, uma lente ocular para visualizar a imagem e uma lente objetiva para ampliar a imagem. Com qualquer técnica de microscopia, é importante entender as partes de um microscópio antes de usá-lo. Em geral, os microscópios compostos usados para imagens de campo claro têm uma ocular na parte superior do osciloscópio, que é presa à cabeça e às objetivas. A ocular tem uma ampliação de 10X e as lentes objetivas são configuradas para uma ampliação específica em uma faixa de 4X-100X. Existem entre três e cinco objetivas em um microscópio padrão. Os objetivos apontam para o palco, que é onde um espécime é colocado para visualização. O palco geralmente tem peças mecânicas e clipes de palco para segurar um slide e movê-lo durante a visualização. Uma abertura é um orifício no palco para a passagem da luz. Esta luz é controlada por uma lente condensadora ajustável acima de um iluminador ou fonte de luz. Para controlar o zoom da platina para o objeto que está sendo visualizado, os microscópios apresentam botões de ajuste de foco grosso e fino. O botão de foco grosseiro se move em uma escala maior do que o foco fino, mas eles estão no mesmo eixo. O foco fino é útil quando o objeto no palco foi aproximado dos objetivos. É importante não deixar a lente objetiva tocar o objeto no palco, pois pode arranhar a lente. Os objetos devem sempre ser visualizados primeiro na objetiva de menor ampliação e claramente focados antes de mudar para objetivas de maior ampliação.
A microscopia é uma ferramenta importante para muitos aspectos da área médica, incluindo pesquisa, diagnóstico e tratamento. Isso tem a aplicação do uso da nanotecnologia na medicina, como um novo método de tratamento no lugar de uma cirurgia mais invasiva5. Os cirurgiões também fazem uso de microscópios, alguns dos quais foram modificados para serem montados na cabeça de um cirurgião e são operados com pedais. Estes são de ampliação muito menor do que até mesmo os microscópios de luz usados hoje, mas facilitam a execução segura de procedimentos delicados, como óptica e neurocirurgia.

Cells are the basic units of life, and their structures help explain how living things work. Cell theory says that every living organism is made of one or more cells, cells are the functional units of organisms, and new cells come from preexisting cells. Cells also share key features, including a plasma membrane, cytoplasm, DNA, and ribosomes.

The plasma membrane is a phospholipid bilayer that surrounds the cell. It separates the cell from its environment and controls what enters and leaves. Inside it, the cytoplasm is a gel-like fluid that contains organic molecules, salts, and other materials needed for cell activity. Many metabolic processes, or life-supporting chemical reactions, take place there.

Cells use DNA as their genetic material, which acts as the blueprint for cellular structures and products. Ribosomes make proteins. Cells are often grouped as prokaryotic or eukaryotic based on where their genetic material is found. Prokaryotes, such as bacteria, have nucleoids instead of a true nucleus, while eukaryotes have membrane-bound nuclei.

Prokaryotic cells are usually small, about 0.1 to 5.0 micrometers, while eukaryotic cells are often 10 to 100 micrometers. Their small size helps prokaryotes move materials quickly and remove waste efficiently. Eukaryotic cells use organelles, such as mitochondria and the Golgi apparatus, to carry out important functions.

Eukaryotic cells are thought to have a shared origin and appear in fossils about 2.4 billion years ago. The earliest eukaryotic cells were larger than prokaryotic cells and likely formed through an endosymbiotic event. In this event, an amoeba-like cell engulfed a micrococcal bacterium, and that bacterium later became the mitochondrion. Mitochondria perform aerobic metabolism and have their own genome.

Mitochondria have two membranes that enclose two compartments. Some reactions that break down high-energy biomolecules happen in the inner compartment. Other reactions in the outer compartment capture released energy and convert it into ATP, the cell’s energy currency. Eukaryotic cells also contain the smooth and rough endoplasmic reticulum, the Golgi apparatus, lysosomes, and vacuoles.

The endoplasmic reticulum means “network inside the plasma,” and it forms a membrane network around the nucleus. Rough ER has ribosomes on its surface and makes proteins, including proteins for the plasma membrane and proteins that are secreted from the cell. Smooth ER makes lipid-based products and helps detoxify harmful chemicals, which is why liver cells contain much of it. Muscle cells also have a lot of smooth ER because it stores calcium needed for contraction.

The Golgi apparatus sorts, modifies, and packages cellular products into vesicles. Some proteins made in the rough ER are digestive enzymes. The Golgi packages these enzymes into lysosomes, which digest food particles taken into the cell and break down old cell parts. Vacuoles are membrane sacs used for storage, including water, metabolic products, or even poisonous molecules, depending on the cell.

Plant and animal cells are both eukaryotic, but they do not have the same organelles. Plant cells have chloroplasts for photosynthesis and often contain chlorophyll, the green pigment. They also have cell walls made of cellulose, which give support and help with water retention. Their larger vacuoles help maintain water pressure, and they also contain plastids that store pigments and products such as starch.

These differences make plant cells and animal cells easy to tell apart. Plant cells usually have a regular, rectangular shape because of their rigid cell walls. Animal cells are more rounded and irregular. Cell walls, chloroplasts, and large vacuoles are key features that distinguish plant cells from animal cells.

Many cells are too small to see without help, so scientists use microscopy to study them. Some cells, such as frog oocytes, are large enough to see with the naked eye, but most samples need magnification. Because many tissues lack color, stains are used to highlight cell parts and tissue types. Methylene blue stains nucleic acids in dead cells by binding to DNA, and safranin stains cell nuclei red.

After staining, cells can be mounted as a wet mount or with immersion oil. In a wet mount, the sample is placed on a slide with liquid between the slide and coverslip. Water or glycerol can be used as the liquid, and glycerol is better for live cultures because it prevents bacterial growth. Immersion oil improves high-magnification viewing because it has the same refractive index as glass and reduces light scattering.

Once stained and mounted, the sample is ready for the microscope. Microscopy methods range from electron scanning technology, which can view objects at atomic scale, to fluorescent live-cell imaging, which tracks molecules in real time. Bright-field microscopy is the simplest method and uses a halogen light source, condenser lens, ocular lens, and objective lens.

Compound microscopes have an eyepiece, a head with objectives, and a stage for the specimen. The eyepiece usually has 10X magnification, and objective lenses commonly range from 4X to 100X. The stage may have clips and mechanical parts to move the slide, and light passes through an aperture in the stage. Coarse and fine focus knobs control sharpness, and the slide should be viewed first at low magnification before moving higher.

Microscopy is important in medicine for research, diagnosis, and treatment. It also supports newer approaches such as nanotechnology in medicine, which can reduce the need for more invasive surgery. Surgeons use modified microscopes as well, including head-mounted models operated with foot pedals. These lower-magnification tools help doctors perform delicate procedures such as optical and neurosurgery safely.
