A extração de DNA é a remoção e purificação do DNA das células. Na impressão digital de DNA, podemos examinar esses fragmentos usando uma técnica chamada eletroforese de DNA ou gel. Para preparar géis, a agarose em pó é misturada com um tampão e aquecida até se dissolver. <h4>Isolando DNA de células</h4>

O isolamento do DNA é um procedimento bastante simples que requer poucas etapas, mas essenciais: coleta de células, lise, degradação de proteínas e, finalmente, precipitação de DNA. Geralmente, o DNA pode ser colhido de pequenas quantidades de tecido, como uma única folha de uma planta, folículos pilosos de animais ou células epiteliais que podem ser facilmente removidas dentro da boca com uma solução salina simples. Como o DNA é compartimentado dentro do núcleo ou mitocôndrias dentro das células eucarióticas, os cientistas primeiro separam a célula e a membrana nuclear para expor o DNA, o que é feito com a etapa de lise. No entanto, uma vez que o DNA é exposto, ele pode ser degradado por íons de metais pesados e extremos de pH. Portanto, uma solução tampão contendo detergente é usada não apenas para dissolver a membrana celular, mas também para proteger o DNA de elementos degradantes na solução. Além disso, o DNA é uma molécula incomumente longa que é condensada dentro do núcleo, compactando-se firmemente em torno de proteínas especiais chamadas histonas. Portanto, uma enzima chamada proteinase K, que degrada as ligações peptídicas das proteínas, é adicionada para separar o DNA das histonas e outras proteínas. Em seguida, os cientistas separam o DNA do resto do extrato celular usando suas propriedades químicas. Devido à estrutura carregada negativamente dos ácidos nucléicos, os íons Na+ em uma solução de cloreto de sódio se ligam e se integram às fitas de DNA emaranhadas. Uma vez que o DNA é agrupado, ele pode ser observado a olho nu. O DNA é insolúvel em álcool e precipita prontamente em baixas temperaturas, portanto, os cientistas adicionam álcool frio para precipitar o DNA. Finalmente, o DNA precipitado pode ser dissolvido e armazenado em água deionizada ou em um tampão suave, livre de quaisquer enzimas que possam degradar os ácidos nucléicos, até que seja usado.

<h4>Quantificando o DNA</h4>

As reações que usam DNA geralmente requerem alta pureza e quantidades precisas de DNA. Portanto, após o isolamento do DNA, os cientistas usam um espectrofotômetro para quantificar a quantidade de DNA em sua amostra, bem como a pureza do DNA na amostra. Um espectrofotômetro mede a intensidade de um feixe de luz passado por uma amostra em função de seu comprimento de onda. Os ácidos nucléicos absorvem luz a 260 nm, enquanto as proteínas absorvem luz a 280 nm. Ao medir a intensidade de um feixe de luz a 260 nm, os cientistas podem determinar a concentração de DNA. Além disso, avaliando a intensidade da luz em 260 nm e 280 nm, a pureza do conteúdo de DNA pode ser determinada. Em uma amostra de DNA puro, a proporção de 260/280 nm deve se aproximar do valor de 2.

<h4>Enzimas de Restrição</h4>

Para uma série de análises, como clonagem, sequenciamento e mapeamento de fragmentos de DNA e genomas, o DNA isolado precisa ser cortado em sequências específicas usando enzimas de restrição. Essas enzimas são produzidas por bactérias como mecanismos de imunidade inata contra a intrusão do DNA viral e funcionam como tesouras moleculares. Assim, depois que as enzimas de restrição identificam um DNA viral, elas começam a separá-lo, cortando-o em segmentos em certas sequências de nucleotídeos que reconhecem. Essas sequências de nucleotídeos geralmente têm de seis a doze nucleotídeos e geralmente são palindrômicas, o que significa que elas têm a mesma sequência de nucleotídeos nas direções 3'-5' e 5'-3'. Por exemplo, a enzima de restrição EcoRI corta o DNA na sequência 3'... GAATTC...-5', que lê o mesmo na direção 5'-3'. Como o EcoRI, existem centenas de enzimas que correspondem a muitas sequências palindrômicas distintas. Uma vez que o DNA é cortado por essas enzimas, os cientistas podem carregar o DNA em um gel em uma câmara de eletroforese e passar corrente elétrica através do gel. O DNA carrega cargas negativas, o que faz com que os fragmentos de DNA migrem em direção ao ânodo. No entanto, os poros do gel retardam fragmentos de DNA maiores em relação a sequências de DNA menores, portanto, as peças se separam de acordo com seu tamanho. Após a conclusão da eletroforese, os fragmentos de DNA podem ser visualizados com o auxílio de corantes que se ligam especificamente às moléculas de DNA.

Cortar DNA com enzimas de restrição e separar fragmentos de DNA com eletroforese permite que os pesquisadores identifiquem os tamanhos de fragmentos desconhecidos em relação a uma escada padrão. Esses fragmentos de interesse podem então ser isolados e ligados a um vetor de plasmídeo para clonagem genética. A engenharia genética e as análises laboratoriais foram imensamente aprimoradas com esses métodos. Com o perfil de DNA, os cientistas são capazes de sequenciar genomas para entender melhor o funcionamento dos organismos e também são capazes de identificar e caracterizar certos genes1.

Recentemente, os kits de análise de ancestralidade de DNA tornaram-se populares, permitindo que os usuários determinem sua linhagem genética, encontrem parentes e até descubram sua disposição genética para várias condições de saúde3. Finalmente, com os avanços da biotecnologia e da engenharia genética, a medicina personalizada está ganhando força entre os pesquisadores clínicos para desenvolver tratamentos usando os dados genéticos de um indivíduo, abrindo caminho para tratar efetivamente os pacientes sem efeitos colaterais prejudiciais4.

DNA extraction, quantification, and cutting are key steps in many biology labs. Scientists use these methods to study genes, prepare DNA for testing, and compare fragments for analysis.

DNA isolation starts with a small sample, such as a leaf, hair follicles, or epithelial cells from the mouth collected with a saline solution. The first steps are cell harvesting, lysis, protein degradation, and DNA precipitation. Lysis breaks open the cell and nuclear membranes so the DNA can be released from the nucleus or mitochondria in eukaryotic cells. A detergent-containing buffer helps dissolve membranes and also protects DNA from degrading conditions such as heavy metal ions and extreme pH.

After the DNA is exposed, Proteinase K is added. This enzyme breaks peptide bonds in proteins and helps separate DNA from histones and other proteins. The DNA is then mixed with sodium chloride, and Na+ ions help the tangled DNA strands clump together because nucleic acids carry negative charges. Cold alcohol is added next, since DNA does not dissolve in alcohol and will precipitate at low temperatures. The DNA can then be dissolved in deionized water or a mild buffer for storage, as long as the solution is free of enzymes that could break it down.

Once DNA is isolated, scientists often measure how much DNA they have and how pure it is. A spectrophotometer does this by sending light through the sample at specific wavelengths. DNA absorbs light at 260 nm, while proteins absorb light at 280 nm. The 260 nm reading shows DNA concentration, and the 260/280 ratio helps estimate purity. In a sample of pure DNA, that ratio should approach 2.

For cloning, sequencing, and mapping DNA fragments or genomes, scientists often need to cut DNA at specific sites. They use restriction enzymes, which are bacterial proteins that act like molecular scissors against invading viral DNA. These enzymes recognize short nucleotide sequences, often six to twelve bases long, and the sequences are usually palindromic, meaning they read the same in both directions. EcoRI is one example and cuts at the sequence GAATTC.

After cutting, the DNA fragments are loaded into a gel and separated by electrophoresis. DNA is negatively charged, so it moves toward the anode when an electric current is applied. Smaller fragments travel faster through the gel pores than larger ones, so the pieces separate by size. Dyes that bind to DNA make the fragments visible, and researchers compare them with a standard ladder to estimate fragment size. The fragments can then be isolated and ligated into a plasmid vector for genetic cloning.

These methods support many modern applications. Scientists use DNA profiling to sequence genomes and to identify and study genes. DNA analysis also helps in forensics for criminal and civil cases, in paternity testing, and in checks for genetic diseases. Ancestry kits use DNA to help people trace lineage, find relatives, and learn about possible health traits. These tools also support personalized medicine, where researchers use a person’s genetic data to design treatments with fewer harmful side effects.
