Enzymes and Activation Energy

Em uma reação química, a energia de ativação, ou Ea, é a diferença de energia livre entre os substratos em seu estado fundamental e em seu estado de transição de alta energia, uma estrutura transitória instável.
Esses reagentes devem ter energia cinética suficiente para colidir com frequência e sofrer modificações químicas para formar produtos.
Devido à energia de ativação de uma reação, o estado de transição tem uma energia maior do que os reagentes ou os produtos. Ele pode perder rapidamente essa energia e retornar aos reagentes ou se converter em produtos.
Quando os substratos se ligam às suas enzimas, a energia de ativação diminui, permitindo que mais reagentes atinjam o estado de transição. Isso permite que mais moléculas sejam transformadas em produtos e aumenta a taxa de uma reação catalisada por enzima em comparação com uma não catalisada.

Como são ligações que armazenam energia, elas liberam energia quando quebradas. No entanto, a molécula deve ser um pouco distorcida para chegar a um estado que permita a quebra das ligações. Uma pequena entrada de energia é necessária para atingir esse estado instável e de alta energia, chamado estado de transição. Por essa razão, as moléculas reagentes não duram muito em seu estado de transição, mas prosseguem muito rapidamente para as próximas etapas da reação química. O estado de transição da reação existe em um estado de energia superior ao dos reagentes e, portanto, Ea é sempre positivo. how enzymes lower reaction energy

Enzymes lower the energy needed for a chemical reaction to begin. That starting energy is called activation energy, or free energy of activation, and it is often written as Ea. During a reaction, bonds break and new bonds form. In glucose breakdown, for example, bonds between carbon atoms break and release energy because they are energy-storing bonds.

Before a reaction can move forward, reactant molecules must reach a high-energy, unstable shape called the transition state. To get there, the molecules may need to become somewhat contorted. This small energy input is required because the transition state sits at a higher energy level than the reactants. For that reason, activation energy is always positive, and reactants do not stay in the transition state for long.

Enzymes work by binding their substrates and forming an enzyme-substrate complex. This complex helps lower activation energy and speeds up the reaction. In some cases, the enzyme shapes the substrate so bond-breaking becomes easier and the transition state is reached more quickly. In other cases, amino acid residues in the enzyme take part in the reaction by providing specific ions or chemical groups that form covalent bonds with the substrate.

After the reaction finishes, the enzyme releases its product or products. The enzyme returns to its original state and is not permanently changed by the reaction it catalyzes. That unchanged recycling is one of the key features that makes enzymes effective catalysts.
