Ribozymes

As ribozimas são tipos especiais de RNA que podem atuar como enzimas. Os substratos para as ribozimas mais naturais são ligações de fosfodiésteres RNA. ribozimas

O termo ribozima é usado para RNA que pode atuar como uma enzima. As ribozimas são encontradas principalmente em vírus, bactérias, organelas vegetais e eucariotos inferiores selecionados. As ribozimas foram descobertas pela primeira vez em 1982, quando o laboratório de Tom Cech observou íntrons do Grupo I agindo como enzimas. Isto foi logo seguido pela descoberta de outra ribozima, Ribonulcease P, pelo laboratório de Sid Altman. Tanto Cech quanto Altman receberam o Prêmio Nobel de Química em 1989 por seu trabalho com ribozimas. As ribozimas podem ser categorizadas em dois grupos dependendo do seu tamanho – grandes e pequenos. As ribozimas grandes podem variar em tamanho de algumas centenas a vários milhares de nucleotídeos. Os íntrons tipo I e II e a ribonuclease P bacteriana são ribozimas grandes. As ribozimas pequenas têm de 30 a 150 nucleotídeos de comprimento. A maioria das ribozimas que ocorrem naturalmente catalisam a autoclivagem, quebrando as ligações fosfodiéster presentes em seu próprio RNA. Ao contrário de uma enzima proteica típica, a maioria das ribozimas realiza uma reação de renovação única porque, após a autoclivagem, elas não estão mais ativas. No entanto, duas ribozimas - Ribonulcease P e o RNA 23S na subunidade ribossômica 50S realizam reações diferentes. A ribonuclease P bacteriana é um complexo RNA-proteína que possui atividade endonuclease e requer íons Mg2+. Seu componente de RNA atua na extremidade 5' do tRNA prematuro para produzir a extremidade 5' madura. Porém, com a evolução de formas de vida complexas, proteínas com vinte aminoácidos podem ter começado a atuar como enzimas e assumido as diversas reações realizadas pelas ribozimas. Esta teoria obtém apoio das ribozimas artificiais desenvolvidas in vitro que podem realizar uma infinidade de reações, como formação de ligações amida, formação de ligações glicosídicas, formação de ligações carbono-carbono e reações de oxidação-redução.

Ribozymes are RNA molecules that can act as enzymes. They help speed up specific chemical reactions without being protein enzymes. This makes them an important example of RNA with more than one job in the cell.

Some ribozymes can cut RNA molecules, and others can join them together. This shows that RNA can do catalytic work as well as carry genetic information. Ribozymes are a key part of how scientists think about the many roles of RNA in biology.

One well-known ribozyme is ribonuclease P, or RNase P. It helps process precursor tRNA into mature tRNA by removing extra RNA at one end. This process is important because tRNA must be correctly trimmed before it can function in protein synthesis.
