O cloreto de sulfonila reage com aminas primárias e secundárias para produzir sulfonamidas substituídas correspondentes, mas não reage com aminas terciárias, pois a amina não possui prótons N-H.
O grupo sulfonil de retirada de elétrons torna o próton sulfonamida N-H ácido o suficiente para reagir com uma base para produzir um sal solúvel em água.
Para distinguir entre os tipos de aminas, as respectivas misturas de aminas em base aquosa são tratadas com cloreto de benzenossulfonila - também conhecido como reagente de Hinsberg. Isso é chamado de teste de Hinsberg.
Uma amina primária reage com cloreto de benzenossulfonila para formar um precipitado de sulfonamida N-substituído que se dissolve na base para gerar uma solução límpida de um sal solúvel em água. Além disso, a adição de um ácido precipita a sulfonamida N-substituída.

The Hinsberg test identifies primary, secondary, and tertiary amines by their reaction with benzenesulfonyl chloride, also called the Hinsberg reagent. It is named after Oscar Hinsberg. The test uses excess aqueous base, and the mixture is then acidified to show different results for each amine type.

A primary amine reacts with the Hinsberg reagent to form an N-substituted benzenesulfonamide. The sulfonyl group pulls electron density away and makes the N-H proton acidic. Because of this, base can remove that proton and form a water-soluble salt. When the solution is acidified again, the N-substituted benzenesulfonamide precipitates.

A secondary amine gives an insoluble N,N-disubstituted sulfonamide after reaction with benzenesulfonyl chloride. This product has no N-H proton, so it does not react with the base. A tertiary amine does not react with the Hinsberg reagent at first. After acidification, however, the tertiary amine is protonated and becomes water-soluble.
