Turnover Number and Catalytic Efficiency

O número de rotatividade ou kcat indica a rapidez com que uma enzima pode converter moléculas de substrato em produtos.
kcat é igual ao número máximo de moléculas de substrato que podem ser transformadas em um determinado tempo por sítio ativo da enzima.
Os números de renovação de diferentes enzimas variam de menos de uma molécula de substrato a milhões de moléculas por segundo.
Para calcular kcat, a velocidade máxima ou Vmax de uma reação catalisada por enzima é dividida pela concentração total da enzima.
Tanto kcat, a taxa catalítica, quanto KM, a afinidade por um substrato, afetam a eficiência catalítica de uma enzima - a eficácia com que uma enzima acelera uma determinada reação bioquímica.
Uma maneira de medir a eficiência catalítica para um substrato específico é a proporção de kcat para KM.

O número de renovação, ou número de rotatividade, de uma enzima é o número máximo de moléculas de substrato que ela pode transformar por unidade de tempo. Os números de rotatividade para a maioria das enzimas variam de 1 a 1.000 moléculas por segundo. A catalase tem o maior número de rotatividade conhecido, capaz de converter até 2,8×106 moléculas de peróxido de hidrogênio em água e oxigênio por segundo. A lisozima tem o menor número de rotatividade conhecido de meia molécula por segundo. A quimotripsina é uma enzima pancreática que decompõe as proteínas durante a digestão. O número de rotatividade da quimotripsina é de 100 moléculas por segundo. Se esta reação ocorresse sem catalisação, as ligações peptídicas levariam centenas de anos para se quebrar na água em pH neutro. Assim, o alto número de rotatividade da quimotripsina ajuda na rápida digestão das proteínas no intestino. A enzima ribulose 1,5-bifosfato carboxilase oxigenase, ou RuBisCO, tem um número de rotatividade muito baixo de fixação de 3 moléculas de CO2 por segundo e é uma das enzimas mais lentas. how fast enzymes work in digestion

Enzymes can work at very different speeds, and their turnover number shows how fast they convert substrate molecules into products. Turnover number is the maximum number of substrate molecules an enzyme can transform per unit time. For most enzymes, this value is between 1 and 1000 molecules per second.

Some enzymes are especially fast. Catalase has the highest known turnover number and can convert up to 2.8×10^6 molecules of hydrogen peroxide into water and oxygen each second. Lysozyme has the lowest known turnover number, at half a molecule per second.

Digestion also depends on enzyme speed. Chymotrypsin is a pancreatic enzyme that breaks down proteins during digestion, and its turnover number is 100 molecules per second. Without an enzyme, peptide bonds would take hundreds of years to break in water at neutral pH. Chymotrypsin helps make protein digestion in the intestine much faster.

Not every slow enzyme is unimportant. RuBisCO, or ribulose 1,5-bisphosphate carboxylase oxygenase, has a very low turnover number and fixes only 3 molecules of CO2 per second. It is one of the slowest enzymes, but it is very abundant in nature. RuBisCO makes up around 50% of the total protein found in leaves.

A high turnover number does not always mean high catalytic efficiency. Catalytic efficiency is the ratio of turnover number, kcat, to affinity, KM. An enzyme also needs a low KM for its substrate to be efficient. The average catalytic efficiency of most enzymes is about 10^5 M^-1 s^-1, so they are moderately efficient. A few enzymes reach 10^8 to 10^9 M^-1 s^-1 and are called superefficient or catalytically perfect.
