Sucrose-secreting Cyanobacteria

Sucrose-secreting cyanobacteria are photosynthetic microorganisms engineered to produce and release sucrose into their surrounding medium, creating renewable platforms for biological production. Using light energy, they fix carbon dioxide through photosynthesis and redirect a portion of the resulting carbon toward sucrose synthesis; genetic modification can enhance production and introduce or regulate transport systems that export sucrose from the cell. In bioengineering, these strains function as photosynthetic cell factories for generating soluble carbon products, potentially simplifying downstream recovery because the product accumulates outside the biomass. Their study also supports efforts to improve carbon utilization, secretion efficiency, and sustainable production under controlled cultivation conditions.

Here, we present a reliable and easy assay to measure the glycogen content in cyanobacterial cells. The procedure entails precipitation, selectable depolymerization, and the detection of glucose residues. This method is suitable for both wildtype and genetically engineered strains and can facilitate the metabolic engineering of cyanobacteria.

Liquid Phase Reactor: Sucrose Inversion

2023

Source: Kerry M. Dooley and Michael G. Benton, Department of Chemical Engineering, Louisiana State University, Baton Rouge, LA Both batch and continuous flow reactors are used in catalytic reactions. Packed beds, which use solid catalysts and a continuous flow, are the most common configuration. In the absence of an extensive recycle stream, such packed bed reactors are typically modeled as "plug flow". The other most common continuous reactor is a stirred tank, which is assumed to be perfectly...

2025

Source: Lea-Smith, D. J., et al. Generation of Marked and Markerless Mutants in Model Cyanobacterial Species. J. Vis. Exp. (2016)The video demonstrates a method for generating antibiotic-marked mutants in cyanobacteria. The protocol begins with culturing cyanobacteria, followed by centrifugation and washing steps that preserve surface pili. A plasmid carrying an antibiotic resistance gene flanked by homologous sequences is then introduced. During incubation, the plasmid is internalized via pili...

Purification of Bacteriophages Using a Sucrose Density Gradient

Source: Rustad, M., et.al. Synthesis of Infectious Bacteriophages in an E. coli-based Cell-free Expression System. J. Vis. Exp. (2017)This video demonstrates the purification of bacteriophages from a suspension containing host bacterial debris using a sucrose density gradient. It shows how the density gradient separates phages by buoyant density, enabling recovery of a visible band for resuspension and further analysis.
