Microglial Mitochondria

Microglial mitochondria are the energy-producing organelles that support microglial survival, surveillance, and responses to injury in the central nervous system. By regulating oxidative phosphorylation, reactive oxygen species, calcium handling, and mitochondrial dynamics, they help determine whether microglia maintain homeostasis or adopt inflammatory and repair-associated states. In bioengineering, microglial mitochondria are studied using organ-on-a-chip platforms, engineered neural tissues, and metabolic imaging to model neuroinflammation and neuron–glia interactions under controlled conditions. These systems can clarify how mitochondrial dysfunction contributes to neurological disease and support the development of targeted therapies that restore cellular metabolism while limiting harmful inflammation.

Multi-parameter Measurement of the Permeability Transition Pore Opening in Isolated Mouse Heart Mitochondria

A spectrofluorometric protocol for the measurement of the mitochondrial permeability transition pore opening in isolated mouse heart mitochondria is presented here. The assay involves the simultaneous measurement of mitochondria Ca2+ handling, mitochondrial membrane potential and mitochondrial volume. The procedure for obtaining high-quality and functional heart mitochondria is also described.

Purification of Mitochondria from Yeast Cells

We describe a rapid and effective method for purification of mitochondria from the yeast Saccharomyces cerevisiae. This method enables the high-yield isolation of pure mitochondria that are essentially free of contamination by other organelles and retain their structural and functional integrity after their purification.
