Cancer Cell Spheroids

Cancer cell spheroids are three-dimensional aggregates of cancer cells that reproduce key features of solid-tumor architecture more realistically than conventional two-dimensional cultures. They form when cells grow under nonadherent or low-attachment conditions, where cell-cell adhesion and extracellular matrix interactions drive compaction while limited diffusion creates oxygen, nutrient, and drug-penetration gradients. These gradients generate distinct proliferative, quiescent, and sometimes necrotic regions, enabling researchers to study tumor growth, treatment response, invasion, and resistance. In cancer research, spheroids support more predictive drug screening and investigations of the tumor microenvironment, helping bridge findings from cell culture to animal models and potential clinical applications.

A Cancer Cell Spheroid Assay to Assess Invasion in a 3D Setting

This method evaluates cancer cell invasion from spheroids into a surrounding 3D matrix. Spheroids are generated via the hanging drop culture method and then embedded in a matrix comprised of basement membrane materials and type I collagen. Invasion out of the spheroids is subsequently monitored.
