Fluorescent Dna Staining

Fluorescent DNA staining is a technique that labels cellular DNA with fluorescent probes, enabling researchers to visualize and measure genetic material in individual cells. The probes bind to or intercalate with DNA and emit light after excitation at a suitable wavelength; fluorescence intensity and distribution can then be recorded by microscopy or flow cytometry. In cancer research, this method supports nuclear imaging, DNA-content analysis, cell-cycle assessment, and detection of structural changes associated with cell death or malignancy. By providing quantitative and spatial information about DNA, fluorescent staining helps characterize tumor cells, evaluate treatment responses, and compare cellular states across experimental conditions.

A method for fluorescent staining of fixed biological material with the specific DNA label methyl green is described. Methyl green is used in a diluted aqueous solution and is very resistant to photobleaching. Its far-red emission allows for deep specimen imaging, making it particularly adequate for whole embryos.

Extracellular DNA Staining: A Method to Identify Extracellular DNA in FFPE Tissue Sections

This video describes the technique of staining extracellular DNA in FFPE tissue sections. The stained extracellular DNA can be quantitated to determine the efficacy of any therapeutic treatment that leads to the death of cancer cells.

Ethanol Fixation and DAPI Staining: A Method to Visualize DNA in C. elegans

Here, we describe a technique used to visualize genomic DNA in fixed, intact nematodes. The video includes an example protocol to count chromatin in unfertilized C. elegans oocytes.

Assessing DNA Damage Foci: To Identify Radiation-Induced DNA Damage Foci in Cancer Cell Line Using Immunofluorescence Staining

2023

This video describes a protocol to detect radiation-induced DNA repair foci in the colon cancer cell lines using immunofluorescence staining. DNA repair proteins get activated at the DNA damage site; therefore, we can detect them using specific antibodies and visualize them under a fluorescence microscope.

2016

Drosophila oogenesis continues to be exceptionally useful in the study of mitochondrial proliferation and inheritance. This manuscript describes a detailed protocol used to label the replicating mitochondrial DNA (mtDNA) in Drosophila adult ovaries with 5-ethynyl-2´-deoxyuridine (EdU), which facilitates uncovering mechanisms associated with mitochondrial inheritance that were previously debatable.
