Absolute Counting Beads

Absolute counting beads are standardized particles used in flow cytometry to determine the number of cells in a sample, rather than only their relative frequency. A known quantity or concentration of fluorescent beads is added to the specimen; after acquisition, the ratio of cellular events to bead events converts gated cell counts into absolute concentrations, commonly cells per volume. In immunology and infection research, this approach quantifies leukocyte subsets, pathogen-infected cells, or immune responses in blood and other specimens. It improves comparisons among samples by accounting for differences in volume and cell recovery, supporting disease monitoring, experimental analysis, and evaluation of treatment-related changes.

Here we present addressable droplet microarrays (ADMs), a droplet array based method able to determine absolute protein abundance in single cells. We demonstrate the capability of ADMs to characterize the heterogeneity in expression of the tumor suppressor protein p53 in a human cancer cell line.

Using a Hemacytometer to Count Cells

2023

Many biomedical experiments require manipulation of a known quantity of cells, in order to achieve accurate, reproducible, and statistically-relevant data. Therefore, learning how to count cells is a particularly essential technique for any successful biomedical scientist. The most common way to count cells is by using a hemacytometer - an instrument that bears two laser-etched grids, which aid in the enumeration of an aliquot cells under a simple light microscope. This data can then be used to...

Mean Absolute Deviation

The mean absolute deviation is also a measure of the variability of data in a sample. It is the absolute value of the average difference between the data values and the mean. Let us consider a dataset containing the number of unsold cupcakes in five shops: 10, 15, 8, 7, and 10. Initially, calculate the sample mean. Then calculate the deviation, or the difference, between each data value and the mean. Next, the absolute values of these deviations are added and divided by the sample size to...

Determining the number of cells in culture is important in standardization of culture conditions and in performing accurate quantitation experiments. In this video, we demonstrate how cells are counted using a hemacytometer.

Absolute Value Inequalities

The absolute value is a mathematical tool that represents the distance of a number from zero on the number line, regardless of its sign. In the context of inequalities, absolute value expressions help define a range of permissible values or boundaries for a variable. These inequalities are commonly used in scientific modeling and data interpretation, where variability within or beyond a certain threshold must be captured precisely. An absolute value inequality of the form ∣x∣ ≤ a, where a ≥ 0,...
