Viral Production

Viral production is the controlled generation and amplification of viruses, typically by infecting susceptible host cells, for research, diagnostic, therapeutic, or vaccine-related use. It works by providing permissive cells with the conditions needed for viral entry, genome replication, synthesis of viral proteins, assembly of new particles, and their release or recovery from the culture system. In immunology and infection research, produced virus supports studies of host-pathogen interactions, immune recognition, pathogenesis, and antiviral responses. Standardized production and quality assessment help researchers obtain reproducible viral preparations while limiting unwanted variability, making the process important for experimental models and the development of vaccines and antiviral strategies.

Dissecting Innate Immune Signaling in Viral Evasion of Cytokine Production

We describe a protocol to measure the antiviral cytokine production in mice infected with a model herpesvirus, murine gamma herpesvirus 68 (γHV68) that is closely-related to human Kaposi’s sarcoma-associated herpesvirus (KSHV) and Epstein-Barr virus (EBV). Utilizing genetically modified mouse strains and mouse embryonic fibroblasts (MEFs), we assessed the antiviral cytokine production both in vivo and ex vivo. “Reconstituting” the expression of innate immune components in knockout embryonic...

(2014)This video demonstrates the process of infecting murine fibroblast cells with herpesvirus and monitoring the resulting antiviral cytokine release and gene expression. It outlines key steps, including viral infection, media collection for cytokine quantification, and cell harvesting for gene expression analysis.

Source: Eggers, C. H. Phage-Mediated Genetic Manipulation of the Lyme Disease Spirochete Borrelia burgdorferi. J. Vis. Exp. (2022)This video demonstrates the preparation of a genetically modified Borrelia burgdorferi culture engineered with dormant bacteriophage DNA carrying an antibiotic resistance gene. Following antibiotic selection and incubation, a chemical inducer triggers DNA damage, leading to excision, replication, and assembly of new virions containing the resistance gene. After a...

The Virochip is a pan-viral microarray designed to simultaneously detect all known viruses as well as novel viruses on the basis of conserved sequence homology. Here we demonstrate how to run a Virochip assay to analyze clinical samples for the presence of both known and unknown viruses.
