Anterior Segment Isolation

Anterior segment isolation is an ex vivo technique that separates the front portion of the eye from the posterior segment while preserving structures involved in aqueous humor dynamics. In a typical preparation, the cornea, sclera, trabecular meshwork, iris, and ciliary body are dissected and mounted in a perfusion chamber, where culture medium maintains tissue viability and controlled pressure permits measurement of fluid outflow. This model supports investigation of intraocular pressure regulation, glaucoma-related changes in the trabecular meshwork, ocular drug delivery, and anterior-segment biomechanics. By providing a controlled system that retains native tissue architecture, it complements cell culture and in vivo studies in ophthalmic research.

Open globe eye injuries may go untreated for multiple days in rural or military-relevant scenarios, resulting in blindness. Therapeutics are needed to minimize loss of vision. Here, we detail an organ culture open globe injury model. With this model, potential therapeutics for stabilizing these injuries can be properly evaluated.

This video describes the generation of a colorectal mouse model by segmental adeno-cre infection. The infected colon segment leads to tumorigenesis within this segment of the mucosa, resulting in adenomas ultimately progressing to invasive and metastatic carcinoma. This model is an attractive platform for studies of cancer biology and preclinical therapeutic trials.
