Snap Frozen Preparation

Snap frozen preparation is a rapid method for preserving freshly collected tissue by freezing it immediately, helping maintain cellular structure, proteins, nucleic acids, and other molecular features for medical analysis. The sample is quickly cooled to very low temperatures, often using liquid nitrogen or a chilled embedding medium, which limits enzymatic activity and reduces ice-crystal formation when performed correctly. Frozen tissue can then be sectioned with a cryostat for intraoperative diagnosis, histological evaluation, immunostaining, or fluorescence-based analysis. This approach is valuable in pathology and biomedical research because it supports time-sensitive clinical decisions and enables molecular testing that may be compromised by chemical fixation or prolonged processing.

To preserve neuronal processes for ultrastructural analysis, we describe a protocol for plating of primary neurons on electron microscopy grids followed by flash freezing, yielding samples suspended in a layer of vitreous ice. These samples can be examined with a cryo-electron microscope to visualize structures at the nanometer scale.
