Osmium Tetroxide Contrast

Osmium tetroxide contrast is a microscopy method that uses osmium tetroxide to increase the visibility of cellular structures, particularly membranes, in ultrastructural samples. The reagent reacts with and stabilizes unsaturated lipids, depositing electron-dense osmium that preserves membrane architecture and improves contrast for transmission electron microscopy. In neuroscience, this approach supports visualization of myelin, axonal membranes, synaptic terminals, and other fine features of nervous tissue. By revealing cellular organization at nanometer-scale resolution, osmium-based contrast helps researchers assess neural connectivity, tissue integrity, and structural changes associated with development, injury, or disease.

Here, we present a protocol to inject ultrasound microbubble contrast agents into living, isolated late-gestation stage murine embryos.
