Subcutaneous Fat Dissection

Subcutaneous fat dissection is the controlled separation and collection of adipose tissue located beneath the skin, a tissue that stores energy and actively influences metabolism through lipid turnover and signaling. During the procedure, the skin and connective tissue are carefully separated to expose subcutaneous fat, which is excised while minimizing contamination from muscle, fascia, blood, or overlying skin. In biochemistry, the isolated tissue can be weighed, processed, and analyzed for lipid composition, enzyme activity, gene or protein expression, and metabolic responses. These measurements help compare adipose depots and clarify how fat storage, mobilization, and endocrine function contribute to health and disease.

We describe an ovarian fad pad transplantation assay suitable for studies of normal and transformed epithelia of the female reproductive tract. The mouse fat pad allows transplantation of large tissue fragments, is easily accessible for surgery and imaging, and provides the most favorable native environment for tissues of Müllerian origin.
