Muscle Pouch Model

The Muscle Pouch Model is an experimental bioengineering system that places a biomaterial, cell-laden construct, or tissue graft within a surgically created muscle pouch to assess its behavior in a living tissue environment. The surrounding muscle provides mechanical support and access to host cells and blood vessels, enabling researchers to examine integration, vascularization, inflammation, and tissue remodeling under controlled conditions. This model supports the evaluation of scaffolds, engineered muscle, drug-delivery systems, and regenerative therapies before more advanced preclinical studies. By linking material design with biological performance, it helps identify factors that promote functional tissue repair and improve the translation of engineered constructs.

Developing an Experimental Aneurysm Model in a Rabbit Using an Elastase-Treated Vessel Pouch

2025

Source: Wanderer, S., et al. Arterial Pouch Microsurgical Bifurcation Aneurysm Model in the Rabbit. J. Vis. Exp. (2020)This video demonstrates the creation of an experimental aneurysm model in a rabbit using an elastase-treated vessel pouch. After excising a segment of the right CCA, the pouch is treated with elastase, then sutured to the left CCA to restore blood flow. The resulting stress on the weakened pouch walls induces aneurysm formation.

An Air Pouch Mouse Model for Lipopolysaccharide-Induced Inflammatory Exudate Collection

This video describes the procedure for generating an air pouch in the subcutaneous tissue of mice and collecting inflammatory exudates in response to lipopolysaccharide administration in vivo. This video also covers the analysis of the neutrophil infiltration in the air pouch.

An optimized technique for the microsurgical creation of arterial bifurcation aneurysms mimicking bifurcation human cerebral aneurysms is described. A venous pouch is sutured into an artificially created true bifurcation of both common carotid arteries. Facilitated microsurgical techniques and aggressive postoperative anticoagulation and analgesia lead to minimized morbidity rates and high aneurysm patency rates.

Developing and testing endovascular devices for intracranial aneurysm treatment is still of great importance. Most aneurysm models used today miss either the important characteristics of an arterial degenerated wall or the hemodynamics of a true bifurcation. Therefore, we aimed to design a novel arterial pouch bifurcation model in rabbits.

2012

A murine model of neuromuscular electrical stimulation (NMES), a safe and inexpensive clinical modality, to the anterior compartment muscles is described. This model has the advantage of modifying a readily available clinical device for the purpose of eliciting targeted and specific muscle contractions in mice.
