Stage 15-16

Stage 15-16 denotes a defined embryonic developmental window identified by characteristic morphological landmarks, providing a reproducible reference for studying how tissues and organs form. Rather than relying only on elapsed time, researchers assign embryos to these stages by comparing features such as tissue organization, body patterning, and developmental progression within the relevant staging system. In bioengineering, accurate staging helps standardize embryo selection before imaging, microsurgery, biomaterial delivery, genetic manipulation, or tissue analysis. Consistent developmental timing improves experimental reproducibility, supports comparisons across samples, and enables researchers to relate engineered interventions to specific biological processes and outcomes.

This video demonstrates a protocol to enrich thymic epithelial cells (TECs) with density gradient for FACS isolation. It also shows the use of EAK16-II/EAKIIH6 peptides to promote the TEC aggregate formation. The microenvironments of EAK16-II/EAKIIH6 hydrogel provide the 3-D configuration necessary to maintain the survival and function of the TECs.

We present a variation of the QUICK (QUantitative Immunoprecipitation Combined with Knockdown) approach that was introduced previously to distinguish between true and false protein-protein interactions. Our approach is based on 15N metabolic labeling, the modulation of affinities of protein-protein interactions by the presence/absence of ATP, immunoprecipitation, and quantitative mass spectrometry.
