Spectrophotometer Od 540

Spectrophotometer OD 540 is a measurement of optical density, or absorbance, recorded at 540 nanometers with a spectrophotometer to estimate how strongly a biological sample interacts with light at that wavelength. The instrument passes 540-nanometer light through the sample and compares transmitted intensity with a reference, converting the difference into an optical-density value; in turbid suspensions, scattered light also contributes to the reading. In biology, OD540 supports quantitative monitoring of cell or particle concentration, pigment production, and assay signals when the measured range is appropriately calibrated. Replicate readings, blanks, and dilution checks improve reliability and help distinguish biological changes from instrument or sample artifacts.

This communication presents data on the accuracy and reproducibility of the BioSpec-nano UV-VIS spectrophotometer for dsDNA and protein quantitation. Even with ultra-small volumes (1 to 2 L), reproducibility is excellent, while the automated wiping function improves throughput and results in minimal carryover for more precise results.

Microvolume samples are quantified by a spectrophotometer system that uses natural surface tension to retain samples without the use of cuvettes or capillaries. The dynamic range of protein concentrations and speed by which they can be measured are greatly increased with this method.
