Protein-protein Interaction Assay

A protein-protein interaction assay is a laboratory method used to detect, measure, or characterize physical associations between proteins, which can reveal how cellular pathways are regulated. Depending on the format, the assay brings candidate proteins into a defined system and detects binding through reporter signals, affinity capture, fluorescence, or changes in proximity. In cancer research, these assays help map signaling networks, identify interactions that promote tumor growth or treatment resistance, and validate potential drug targets. Comparing interaction strength under different conditions can also clarify how mutations, post-translational modifications, or therapeutic compounds alter cancer-related protein complexes.

Proximity Ligation Assay to Study In Situ Protein-Protein Interactions

This video describes the proximity ligation assay to study in situ protein-protein interactions in nematode cells. The close proximity of the proteins of interest generates the circular DNA template with the help of a PLA probe attached to a secondary antibody. The probe is further amplified using the amplification solution, which produces a long single-stranded DNA followed by the hybridization of the detector probe, enabling the localization of protein-protein interactions in cells.
