Protein Mass Determination

Protein mass determination is the measurement or estimation of a protein’s molecular mass, an essential property for identifying, characterizing, and assessing protein purity in chemistry and biochemistry. In mass spectrometry, proteins are ionized into charged species, their mass-to-charge ratios are measured, and the resulting charge-state pattern is mathematically converted into molecular mass; SDS-PAGE provides an alternative estimate by separating denatured proteins according to size relative to standards. These methods can reveal intact protein mass, truncations, processing, and some chemical modifications. Protein mass determination therefore supports structural analysis, quality control, biomarker research, and the development of biopharmaceuticals.

Determining the Mass Percent Composition in an Aqueous Solution

2023

Source: Laboratory of Dr. Neal Abrams — SUNY College of Environmental Science and Forestry Determining the composition of a solution is an important analytical and forensic technique. When solutions are made with water, they are referred to as being aqueous, or containing water. The primary component of a solution is referred to as the solvent, and the dissolved minor component is called the solute. The solute is dissolved in the solvent to make a solution. Water is the most common solvent in...

Photometric Protein Determination

2023

Measuring the concentration is a fundamental step of many biochemical assays. Photometric protein determination takes advantage of the fact that the more a sample contains light-absorbing substances, the less the light will transmit through it. Since the relationship between concentration and absorption is linear, this phenomenon can be used to measure the concentration in samples where it is unknown. This video describes the basics of photometric protein determination and introduces the...

2010

Mass spectrometry has proven to be a valuable tool for analyzing large protein complexes. This method enables insights into the composition, stoichiometry and overall architecture of multi-subunit assemblies. Here, we describe, step-by-step, how to perform a structural mass spectrometry analysis, and characterize macromolecular structures.

Determination of Molar Masses of Polymers I

2026

Polymerization produces macromolecules with a range of chain lengths due to the random nature of molecular growth processes. As chains form and terminate at different stages, a single polymer sample contains molecules of varying sizes rather than a uniform structure. This variability is described using average molar masses and distribution-related parameters, which together provide a comprehensive understanding of polymer characteristics. The distribution of molar masses plays a critical role in...

Determination of Molar Masses of Polymers II

Polymer samples typically consist of macromolecular chains with a distribution of lengths, resulting in a range of molar masses rather than a single discrete value. Conventional descriptors such as the number-average molar mass and weight-average molar mass quantify this distribution but do not fully capture polymer behavior in solution..The viscosity-average molar mass provides a more realistic description of polymer behavior in solution because it accounts for the enhanced contribution of...
