Pancreatic Differentiation

Pancreatic differentiation is the developmental process through which embryonic progenitor cells become the specialized endocrine and exocrine cells that form the pancreas. During development, signals acting on definitive endoderm establish pancreatic progenitors, while transcription factors including PDX1, NKX6-1, and NEUROG3 guide lineage commitment toward ductal, acinar, or hormone-producing cells. In developmental biology, studying this process reveals how cell fate decisions generate pancreatic tissue and organize its functions. This knowledge supports stem-cell differentiation protocols, disease modeling, diabetes research, drug testing, and efforts to produce insulin-producing cells for regenerative medicine.

Here we describe a 4-stage protocol to differentiate human embryonic stem cells to NKX6-1+ pancreatic progenitors in vitro. This protocol can be applied to a variety of human pluripotent stem cell lines.

Protocols to generate hPSC mutant lines using the iCRISPR platform and to differentiate hPSCs into glucose-responsive β-like cells are described. Combining genome editing technology with hPSC-directed differentiation provides a powerful platform for the systematic analysis of the role of lineage determinants in human development and disease progression.

The current study describes a directed differentiation approach in inducing pancreatic differentiation of human embryonic stem cells. Of great significance is the finding that endothelial cell co-culture mediates maturation of human embryonic stem cell derived pancreatic progenitors into insulin expressing cells.
