Transient Transgenesis

Transient transgenesis is a genetic engineering approach that produces temporary expression of an introduced gene without establishing a heritable change in the genome. Typically, DNA, messenger RNA, or other nucleic acids are delivered into cells or embryos by methods such as microinjection, electroporation, or transfection; the introduced material is then expressed before it is degraded, diluted through cell division, or lost during development. In developmental biology, this technique enables rapid testing of gene function, promoter activity, lineage behavior, and developmental timing using fluorescent or other reporter proteins. It is especially valuable for short-term experiments when generating stable transgenic lines is impractical, although expression can vary among cells and developmental stages.

Transgenic manipulations and genome editing are critical for functionally testing the roles of genes and cis-regulatory elements. Here a detailed microinjection protocol for the generation of genomic modifications (including Tol2-mediated fluorescent reporter transgene constructs, TALENs, and CRISPRs) is presented for the emergent model fish, the threespine stickleback.

In model organisms, transgenesis can manipulate gene functions while RNAi can knockdown specific mRNA transcripts 1-2. This protocol aims to illustrate the techniques needed to introduce stably transmitted DNA and transient double stranded RNA into the necromenic nematode Pristionchus pacificus for studies in evolutionary, developmental, and behavioral biology.

Series R—L Circuit Transients

2024

In a series resistor-inductor (R-L) circuit, closing the switch at the start of the time period simulates a three-phase short circuit, a fault condition where all three phases of an unloaded synchronous machine are short-circuited. When there is no fault impedance and no initial current, the initial voltage is determined by the phase angle of the source voltage. Using Kirchhoff's Voltage Law (KVL) to analyze this circuit helps determine the total asymmetrical fault current, which consists of...
