Yeast Estrogen Screen

The Yeast Estrogen Screen is a cell-based bioassay that detects estrogenic activity in chemicals and environmental samples, supporting the assessment of endocrine-disrupting compounds. In this system, genetically modified yeast express a human estrogen receptor linked to a reporter gene; when an estrogenic substance binds the receptor, it activates gene expression and produces a measurable signal, often through color development. The assay provides a relatively simple, sensitive, and reproducible method for screening wastewater, industrial chemicals, pesticides, and other environmental contaminants. Results can help prioritize substances for further toxicological testing and clarify how chemical pollution may affect hormone-regulated processes in wildlife and humans.

This video describes a cell-based reporter assay to determine the estrogenic activity of plant-derived compounds. A compound with estrogen-like activity mimics its binding to estrogen receptors and drives downstream reporter enzyme gene transcription. The reporter enzyme generates bioluminescence in a reaction, indicating the estrogenic activity of the compound.

Endocrine Disrupting Compounds (EDC) pose a substantial risk to the aquatic environment. Municipal wastewater treatment plants are major contributors to the estrogenic potency of surface waters. The methodology provided in this paper allows for an assessment of the efficacy and suitability of wastewater treatment processes with respect to EDC removal.
