Dissolved Oxygen Detection

Dissolved oxygen detection measures the concentration of oxygen gas present in water, an essential indicator of aquatic ecosystem health and water quality. Electrochemical sensors determine oxygen levels by reducing oxygen at a cathode and relating the resulting current to concentration, while optical sensors measure oxygen-dependent quenching of a luminescent dye. These measurements help assess wastewater treatment, pollution, eutrophication, and the capacity of rivers, lakes, and aquaculture systems to support aerobic organisms. Reliable detection also supports environmental monitoring by revealing changes caused by temperature, salinity, microbial respiration, and chemical oxygen demand.

This article describes a simple and reproducible protocol to manipulate dissolved oxygen conditions in a laboratory setting for animal behavior studies. This protocol may be used in both teaching and research laboratory settings to evaluate organismal response of macroinvertebrates, fishes, or amphibians to changes in dissolved oxygen concentration.

We present an experimental procedure for measuring the partial pressure of oxygen (pO2) in cerebral vasculature based on oxygen-dependent quenching of phosphorescence. Animal preparation and imaging procedures were outlined for both large field of view CCD-based imaging of pO2 in rats and 2-photon excitation based imaging of pO2 in mice.
