C Elegans Microinjection

C. elegans microinjection is a technique for delivering genetic material directly into the gonad of the nematode Caenorhabditis elegans, enabling controlled manipulation of gene expression and inheritance. By immobilizing an adult hermaphrodite and penetrating the syncytial gonad with a fine needle, researchers introduce DNA, RNA, or genome-editing reagents; injected DNA can form heritable extrachromosomal arrays in progeny, while CRISPR-based mixtures can support targeted sequence changes. In genetics, the method enables transgene expression, fluorescent reporter creation, gene rescue, and functional analysis, providing rapid tests of gene function and regulatory elements in vivo.
