Multiplex Reactions

Multiplex reactions are laboratory assays that analyze multiple genetic targets simultaneously within a single reaction, conserving sample, reagents, and time. In multiplex PCR, primer pairs selectively amplify different DNA sequences under shared cycling conditions, while distinct amplicon sizes, fluorescent labels, or sequence-specific probes enable target discrimination; careful primer design and reaction optimization reduce interference and nonspecific amplification. In genetics, these reactions support genotyping, mutation and pathogen detection, linkage analysis, and preparation for high-throughput sequencing. By increasing information obtained from limited samples, multiplexing improves experimental efficiency and enables broader analysis of genetic variation.

Differential diagnostics in painful arthroplasty is crucial for treatment success. Joint aspiration is routinely performed preoperatively. Multiplex polymerase chain reaction of the joint aspirate and the sonication fluid, a feasible tool for quick pathogen detection from these samples is described.

This protocol demonstrates the pan-lyssavirus LN34 real-time reverse transcription-polymerase chain reaction (RT-PCR) assay from tissue collection to result interpretation, including updates to primer sequences and formulations to improve assay performance for some non-rabies lyssaviruses and lagomorphs. We also demonstrate the assay setup for a single-well LN34 multiplexed (LN34M) format.

Multiplex fluorescence immunoassays are becoming a reference for serology testing. Their application, however, is limited by the complexity and time-consuming bead-analyte coupling processes. This report presents a semi-automated, reproducible multiplex microsphere bead coupling protocol that is comparable to the standard method while considerably reducing time to results.
