3d Organ Culture

3D organ culture is a laboratory method that maintains cells or tissue fragments in a three-dimensional arrangement, preserving spatial relationships that are often lost in conventional two-dimensional cultures. Cells grow within a supportive matrix or tissue scaffold, allowing cell-cell interactions, extracellular signaling, and tissue-specific responses to develop under controlled conditions. In immunology and infection research, these models help investigators examine how pathogens interact with host tissues and how immune cells respond within a more realistic microenvironment. They can support studies of infection mechanisms, inflammatory responses, barrier function, and therapeutic interventions while reducing reliance on simplified cell monolayers.

Whole Organ Tissue Culture

Whole organs can be cultured ex vivo using specialized bioreactors with the goal of repairing or replacing entire organs. This method uses a donor organ that is stripped of all cells, leaving behind the three-dimensional structure, which is then repopulated with new cells. This video demonstrates the whole organ culture of lungs and shows how a dynamic culture that mimics the mechanical stimulation in the body is needed to induce native tissue properties.

This article presents a method to fluorescently label collagen that can be further used for both fix and live imaging of 3D cell cultures. We also provide an optimized protocol to visualize endogenous cytoskeletal proteins of cells cultured in 3D environments.
