Common Dendritic Progenitors

Common dendritic progenitors are bone marrow hematopoietic progenitor cells that generate much of the dendritic cell lineage, making them central to the development of innate and adaptive immune responses. They arise from multipotent progenitors and differentiate into conventional dendritic cells and plasmacytoid dendritic cells through coordinated cytokine signaling, particularly Flt3 ligand, and lineage-defining transcription factors such as IRF8 and GATA2. Studying these progenitors clarifies how antigen-presenting cells are produced, specialized, and maintained during infection and inflammation. Their characterization supports research into immune development, pathogen defense, cancer immunology, autoimmune disease, and strategies for modulating dendritic cell responses.

In Vitro Generation of Plasmacytoid Dendritic Cells from Common Lymphoid Progenitors

This video demonstrates an in vitro assay to generate plasmacytoid dendritic cells (pDCs) from common lymphoid progenitor cells (CLPs). Upon co-culturing purified CLPs with γ-irradiated feeder cells in the presence of Fms-like tyrosine kinase-3 ligand (FL), the CLPs differentiate into non-adherent pDCs, the presence of which is confirmed using flow cytometry.
