Il-2 Il-7 Stimulation

IL-2 and IL-7 stimulation is an experimental approach that uses two immune-signaling cytokines to promote the survival, proliferation, and functional maintenance of lymphocytes in culture. IL-2 binds the IL-2 receptor, while IL-7 engages the IL-7 receptor; receptor activation triggers intracellular JAK-STAT signaling and regulates genes involved in cell-cycle progression and survival. In immunology and infection research, this stimulation supports the expansion and characterization of T-cell populations, helps assess cytokine responsiveness, and can improve recovery of lymphocytes from experimental samples. Comparing responses to IL-2, IL-7, or both can clarify how distinct growth signals shape immune-cell persistence and activity.

Here, we present a protocol to detect total cellular reactive oxygen species (ROS) using 2',7'-dichlorodihydrofluorescein diacetate (DCFH-DA). This method can visualize cellular ROS localization in adherent cells with a fluorescence microscope and quantify ROS intensity with a fluorescence plate reader. This protocol is simple, efficient and cost-effective.
