Immunofluorescence Method

The immunofluorescence method is a microscopy-based technique that uses fluorescently labeled antibodies to locate and visualize specific antigens in cells, tissues, or microorganisms. Samples are typically fixed and, when necessary, permeabilized before primary antibodies bind target molecules; fluorescent secondary antibodies can then amplify the signal, which is detected by fluorescence microscopy. In immunology and infection research, immunofluorescence helps identify pathogens, map host proteins, and examine antigen distribution, cellular localization, and changes during infection. Multicolor labeling can reveal relationships among several targets, supporting diagnosis, studies of immune responses, and investigation of pathogen-host interactions.

An In Vivo Immunofluorescence Localization Method to Study Antibody Biodistribution

This video demonstrates an in vivo immunofluorescence localization (IVIL) technique to study the biodistribution of tumor-targeting antibodies in a murine model of breast cancer. Upon the intravenous injection of a tumor-specific antibody in the mouse, the tumor is surgically isolated, and the localization of the antibody within the tumor tissue is confirmed via ex vivo immunostaining.
