Lumen Measurement

Lumen measurement is the quantitative assessment of the inner diameter, cross-sectional area, or volume of a tubular anatomical structure, such as a blood vessel, airway, or gastrointestinal tract. Clinicians obtain these measurements from imaging or direct visualization, including ultrasound, computed tomography, magnetic resonance imaging, and endoscopy, then compare the lumen with expected anatomy or earlier examinations to identify narrowing, enlargement, obstruction, or abnormal remodeling. In medicine, accurate lumen measurement supports diagnosis and severity assessment for conditions such as vascular stenosis, airway disease, and intestinal obstruction. It also helps guide treatment planning, monitor disease progression, and evaluate responses to interventions.

Source: Li, J. et al. Development of a Larval Zebrafish Infection Model for Clostridioides difficile. J. Vis. Exp. (2020)This video demonstrates the microinjection-based delivery of fluorescently labeled Clostridioides difficile into the intestinal lumen of an anesthetized zebrafish larva. It outlines the steps involved in larval preparation, bacterial injection, and fluorescence microscopy–based confirmation of bacterial localization within the intestine.

Source: Patras, K. A. & Doran, K. S. A Murine Model of Group B Streptococcus Vaginal Colonization. J. Vis. Exp. (2016)This video demonstrates the quantification of vaginal Group B Streptococcus (GBS) load in a mouse model using swabbing and chromogenic culture. It outlines the steps involved in sample collection, plating, and colony counting to quantify GBS load in the mouse vaginal lumen.

Gold Immunostaining of Exosomal Sections: A Technique to Visualize Diagnostic Markers in Exosomal Lumen

2023

In this video, we demonstrate gold immunostaining of biomarkers localized within the lumen region of ultrathin exosomal sections. This technique enables early diagnosis of diseases by visualization of target molecules within the exosomes secreted by the diseased cells.

Gut Acidification Monitoring Assay: A Technique to Study the Acidification of the Intestinal Lumen in Drosophila using Bromophenol Blue Dye

2025

This video demonstrates an assay to monitor gut acidification in Drosophila flies. After the ingestion of food supplemented with a pH indicator dye, the acidification of the intestinal lumen results in a change in the color of the dye. Visual inspection of the digestive tract of the flies to examine the color change provides evidence of acidification.

The C. elegans Excretory Canal as a Model for Intracellular Lumen Morphogenesis and In Vivo Polarized Membrane Biogenesis in a Single Cell: labeling by GFP-fusions, RNAi Interaction Screen and Imaging

2017

The C. elegans excretory canal is a unique single-cell model for the visual in vivo analysis of de novo polarized membrane biogenesis. This protocol describes a combination of standard genetic/RNAi and imaging approaches, adaptable for the identification and characterization of molecules directing unicellular tubulogenesis, and apical membrane and lumen biogenesis.
