Bisulfite Sequencing Alternative

A bisulfite sequencing alternative is a DNA methylation profiling method that measures modified cytosines without exposing DNA to bisulfite, which can damage templates and complicate analysis of 5-methylcytosine and 5-hydroxymethylcytosine. Enzymatic methods such as enzymatic methyl-sequencing use TET-mediated oxidation followed by selective cytosine deamination, allowing unmodified cytosines to be read differently from protected methylated bases during sequencing. In neuroscience, these approaches help map methylation and hydroxymethylation across neuronal genomes, including patterns associated with development, synaptic plasticity, aging, and neurological disease. They can improve DNA integrity and provide greater resolution for studying epigenetic regulation in limited or challenging brain samples.
