Microscope Visualization

Microscope visualization is the use of optical or electron-based instruments to resolve structures that are too small to see with the unaided eye, making cellular organization and activity accessible for scientific study. In neuroscience, microscopes form images by directing light or electrons onto a sample and collecting signals produced by transmitted, reflected, or fluorescently labeled components; optical sectioning can isolate details within complex tissue. These approaches reveal neuronal morphology, synaptic connections, and interactions between neurons and glial cells. Quantitative image analysis further supports studies of brain development, circuit organization, disease mechanisms, and responses to experimental treatments.

Source: Brown, A. O., et al. Visualization of Streptococcus pneumoniae within Cardiac Microlesions and Subsequent Cardiac Remodeling. J. Vis. Exp. (2015).This video demonstrates the microscopic visualization of bacterial microlesions in mouse heart tissue. The heart of a mouse infected with Streptococcus pneumoniae is harvested, washed, and fixed before being processed to obtain coronal sections. These sections are stained with hematoxylin and eosin and examined under a microscope to visualize...

Microscopic Visualization of Porous Nanographenes Synthesized through a Combination of Solution and On-Surface Chemistry

2021

A combination of solution and surface assisted synthesis opens new directions in the atomically precise synthesis of nanostructures. Scanning tunneling microscopy (STM) supplemented by non-contact atomic force microscopy (nc-AFM) enables detailed characterization of newly designed and generated carbon-based nano-objects.

Xylem Water Distribution in Woody Plants Visualized with a Cryo-scanning Electron Microscope

Observing the water distribution within the xylem provides significant information regarding water flow dynamics in woody plants. In this study, we demonstrate the practical approach to observe xylem water distribution in situ by using a cryostat and cryo-SEM, which eliminates artifactual changes in the water status during sample preparation.

Major Components of the Light Microscope

2008

The light microscope is a basic tool for the cell biologist, who should have a thorough understanding of how it works, how it should be aligned for different applications, and how it should be maintained as required to obtain maximum image-forming capacity and resolution. The components of the microscope are described in detail here.
