Mito-timer Biosensor

MitoTimer Biosensor is a genetically encoded fluorescent reporter that indicates the relative age and turnover of proteins within mitochondria, making it useful for studying mitochondrial quality in neuroscience. Targeted to mitochondria, the reporter initially emits green fluorescence and progressively shifts toward red as its fluorescent protein chromophore matures and undergoes oxidation, so the red-to-green signal provides a time-dependent measure of mitochondrial protein persistence. Researchers use MitoTimer to examine mitochondrial dynamics, biogenesis, transport, and degradation in neurons, including how aging, injury, or disease alters organelle maintenance. This approach links mitochondrial behavior to neuronal function and pathology.

We describe a rapid and effective method for purification of mitochondria from the yeast Saccharomyces cerevisiae. This method enables the high-yield isolation of pure mitochondria that are essentially free of contamination by other organelles and retain their structural and functional integrity after their purification.
