Polybrene Transduction

Polybrene transduction is a method that uses the cationic polymer polybrene to improve delivery of viral genetic material into cultured cells, including neurons. Polybrene reduces electrostatic repulsion between negatively charged viral particles and cell membranes, increasing virus-to-cell contact and supporting entry during transduction. In neuroscience, this approach commonly enhances delivery by lentiviral or retroviral vectors for stable gene expression, neuronal labeling, or experimental manipulation of neural cells. By improving transduction efficiency, it can help researchers study neuronal development, signaling, connectivity, and disease-related changes, although polybrene concentration and exposure must be optimized because cellular responses can vary.

The goal of this study was to formulate technologies that allow for successful gene transduction in primary natural killer (NK) cells. The dextran-mediated lentiviral transduction of human or mouse primary NK cells results in higher gene expression efficiencies. This method of gene transduction will vastly improve NK cell genetic manipulation.

Enhancing Viral Transduction in Natural Killer Cells Using a Cationic Polymer

2025

This video demonstrates an assay for cationic polymer-mediated viral transduction in natural killer (NK) cells. The cells are incubated with transgenic lentivirus vectors encoding a fluorescent protein. The addition of a cationic polymer enables the virus to attach to the cells and integrate its RNA into the host genome, expressing the fluorescent protein. The fluorescence is quantified to assess the polymer-induced enhancement of transduction.

This manuscript provides a description of the methodology used to establish transduction-transplantation mouse models. A detailed account is given of technical errors to avoid when performing bone marrow transplants. A clear understanding should be gained of the importance of high viral titer, transfection/transduction, and irradiation.

Among the three main modes of HGT—transformation, conjugation, and transduction—transduction is unique in that it is mediated by bacteriophages, or bacterial viruses.Transduction occurs in two ways. Generalized transduction occurs during the lytic cycle of a bacteriophage infection. In this process, bacteriophages infect bacterial cells, replicate within them, and ultimately cause cell lysis, releasing newly assembled virions. Occasionally, random fragments of the bacterial genome are...
