Sucrose Fixation

Sucrose fixation is a tissue-preservation approach that uses sucrose-containing solutions to maintain the structure and molecular accessibility of biological specimens, including nervous tissue. In neuroscience workflows, sucrose treatment often follows chemical fixation and acts through osmotic equilibration, replacing water within tissue and reducing damage from freezing during cryostat sectioning. This preparation helps preserve delicate brain architecture, neuronal processes, and antigenic targets for microscopy, immunohistochemistry, and related analyses. By improving section quality and limiting structural artifacts, sucrose fixation supports reliable visualization of cellular organization, protein distribution, and neural connections in experimental and pathological research.
