This video demonstrates the recording of a direct-coupled electroretinogram (DC-ERG) on a dark-adapted, anesthetized mouse retinal pigment epithelium (RPE). After applying dilating drops, the animal is placed in a recording setup, and the capillary electrode filled with a salt buffer is placed on the cornea. After establishing the baseline, DC-ERG is recorded.

Protocol

All procedures involving animal models have been reviewed by the local institutional animal care committee and the JoVE veterinary review board.

1. Mouse and electrode positioning

  1. Keep the mice overnight in a well-ventilated, light-tight box for dark adaptation.
  2. The animals were anesthetized by intraperitoneal injection of ketamine (80 mg/kg) and xylazine (8 mg/kg).
  3. Apply a drop of 0.5% proparacaine HCl topically to anesthetize the cornea, as well as a drop of 2.5% phenylephrine HCl and 0.5% tropicamide to dilate the pupils.
  4. Trim the mouse whiskers with scissors to prevent inadverte....
