Kronisk okulær hypertension fremkaldes ved hjælp laserfotokoagulation af trabekelværket i muse øjne. Det intraokulære tryk (IOP) er forhøjet i flere måneder efter laserbehandling. Faldet af visuel skarphed og kontrast følsomhed forsøgsdyr overvåges ved hjælp af optomotor testen.
Glaukom, ofte forbundet med forhøjet intraokulært tryk (IOP), er en af de førende årsager til blindhed. Vi søgte at etablere en musemodel af okulær hypertension at efterligne human højspænding glaukom. Her laserbelysning påføres hornhindens limbus at photocoagulate den vandige udstrømning, inducere nævervinklet. Ændringerne i IOP overvåges ved hjælp af en rebound tonometer før og efter laserbehandlingen. En optomotor adfærdsmæssige test anvendes til at måle tilsvarende ændringer i visuel kapacitet. Den repræsentativt resultat fra en mus, der udviklede vedvarende IOP elevation efter laserbelysning vises. En nedsat synsstyrke og kontrast følsomhed er observeret i denne okular hypertensive mus. Sammen vores undersøgelse indfører en værdifuld modelsystem til at undersøge neuronal degeneration og de underliggende molekylære mekanismer i glaucomatøse mus.
Vi rapporterer over at vedvarende okulær hypertension kan induceres ved laserbelysning i muse øjne. Sammenlignet med saltvand injektion model 18 og venen cautery model 11 begge kræver omfattende mikrokirurgiske færdigheder er laserbelysning forholdsvis enkel og let at udføre. Normalt kan vi udføre laserbelysning til 4-6 mus i 2-3 timer. De kritiske skridt til at opnå vedvarende IOP elevation er forkammeret udfladning før laser og parametrene for laserbelysning.
