Protein crystallography is the most successful technique in structural biology, with the vast majority of published structures having been determined using this method. Highly detailed electron density maps are used to model structures of proteins and ligands with vast applications in both academia and industry. In addition to basic scientific and academic investigations, the use of high-throughput approaches within this field is of great benefit to drug discovery and development. Harnessing the full power of fourth-generation synchrotrons, microfocus beamlines, and high frame rate pixel detectors depends on the coevolution of methods in crystal growth, harvesting, data collection, and processing, to keep pace with these developments. Decades-old traditional techniques are increasingly combined with or replaced by new methods, resulting in innovative methodologies with growing sophistication. This collection of video protocols bridges the gap between traditional and state-of-the-art techniques in macromolecular crystallography, providing a useful repository of visual methods that can serve as a reference for established techniques or introducing new practices in an ever-growing scientific field.
