The plant cell wall constitutes a complex network comprising polysaccharides such as cellulose, hemicelluloses, pectins, and structural proteins. It serves as a frontline defense mechanism against environmental and developmental stimuli, acting as a barrier and perceiving stress signal through cell wall receptors. This, in turn, triggers cell wall remodeling and signaling to induce various responses, including defense against diverse stresses. To date, many cell wall components have been identified to impact plant development and tolerance to environmental stresses. However, our understanding of changes in cell wall properties and structures under different stresses remains largely unexplored in the context of recent advances in primary cell wall structure.
The objective of this Methods Collection is to present recent innovative research employing traditional and modern biotechnologies to uncover the intricate structural and dynamic changes of specific wall polymers with improved temporal-spatial resolution. Furthermore, employing advanced microscopic technologies to elucidate the native-state structure of polysaccharides in three dimensions with nanometer precision will significantly deepen our comprehension of plant cell wall structure and function.
We enthusiastically welcome studies focusing on the characterization of novel cell wall components involved in plant development and defense responses. Additionally, investigations elucidating the high supramolecular complexity of the plant cell wall itself, alongside changes in the localization of specific cell wall epitopes and wall composition during various biological events, are highly encouraged.
