Dr. Lin received his PhD degree from Leibniz Institute for Neurobiology (LIN), which approved by Otto-von-Guericke-Universität Magdeburg (OVGU) in 2021, Magdeburg, Germany. In 2022, Dr. Lin started his post-doctoral research in Guangdong Institute of Intelligence Science and Technology, Zhuhai, Guangdong, China. Starting from his PhD work, Dr. Lin was interesting in Calcium signal regulation in the central nerve system, and he focus mainly on calcium channel-related proteins on the cell plasma membrane, by using single cell Calcium imaging in combination with electrical or chemical stimulation, the calcium dynamic properties was characterized. Currently, Dr. Lin started with 2 photo microscopy to investigated calcium regulation in vivo.

Dr. Zhu received her PhD in Biomedical Engineering from the Hong Kong Polytechnic University in July 2022. She has a particular interest in ultrasound neuromodulation and has dedicated her efforts to investigating its mechanisms, exploring the development of sonogenetic tools based on her findings. During her research, she utilized various calcium recording methods, including fiber photometry and in vitro calcium imaging. After joining the Guangdong Institute of Intelligence Science and Technology in Zhuhai, Guangdong, China, in August 2022, she continued to expand her research, incorporating more in vivo calcium imaging techniques such as wide-field calcium imaging, two-photon calcium imaging, and miniscope technology.

Collection Overview

Calcium homeostasis and signal transduction play pivotal roles in the physiology and biochemistry of all organisms. Calcium signaling dysfunction in the central nervous system is associated with numerous neurological diseases, including schizophrenia, autism spectrum disorder, and Alzheimer’s disease. Therefore, characterizing calcium dynamics in neurons, including their organelles and at the brain level, would help understand the molecular mechanisms underlying these neurological diseases.

Numerous calcium detectors have been developed for this purpose, such as genetically encoded calcium indicators from the GCaMP family and chemical compounds from the Fluo family. These calcium detectors have been widely applied in cell culture and mouse models. By combining them with electrophysiology and two-photon microscopy, researchers have observed many exciting phenomena and answered many scientific questions. However, the calcium level in the cytosol is 50 to 100 nM, while in the extracellular space, it is 1 to 2 mM. Selecting an appropriate calcium detector for different brain areas or even different organelles still needs further investigation.

In this Methods Collection, we present a variety of neuron calcium imaging methods in the mammalian nervous system.
